The glucocorticoid receptor and FOXO1 synergistically activate the skeletal muscle atrophy-associated MuRF1 gene.
David Waddell,Leslie M. Baehr,Jens van den Brandt,Steven A. Johnsen,Holger M. Reichardt,J. David Furlow,Sue C. Bodine +6 more
TL;DR: Transient transfection assays demonstrate that the MuRF1 promoter is responsive to both the dexamethasone (DEX)-activated glucocorticoid receptor (GR) and FOXO1, whereas coexpression of GR and FoxO1 leads to a dramatic synergistic increase in reporter gene activity, presenting new insights into the role of the GR andFOXO family of transcription factors in the transcriptional regulation of the Mu RF1 gene.

Fibroblast drug scavenging increases intratumoural gemcitabine accumulation in murine pancreas cancer.
Elisabeth Hessmann,Melanie S. Patzak,Lukas Klein,Nai-Ming Chen,Vijayalakshmi Kari,Iswarya Ramu,Tashinga E. Bapiro,Tashinga E. Bapiro,Kristopher K. Frese,Aarthi Gopinathan,Frances M. Richards,Duncan I. Jodrell,Caroline S. Verbeke,Caroline S. Verbeke,Xuan Li,Rainer Heuchel,Johannes-Matthias Löhr,Steven A. Johnsen,Thomas M. Gress,Volker Ellenrieder,Albrecht Neesse +20 more
TL;DR: It is suggested that fibroblast drug scavenging may contribute to the clinical failure of gemcitabine in desmoplastic PDAC and metabolic targeting of CAFs may be a promising strategy to enhance the antiproliferative effects of gem citabine.

HDAC inhibitor–dependent transcriptome and memory reinstatement in cognitive decline models
Eva Benito,Hendrik Urbanke,Binu Ramachandran,Jonas Barth,Rashi Halder,Ankit Awasthi,Gaurav Jain,Vincenzo Capece,Susanne Burkhardt,Magdalena Navarro-Sala,Sankari Nagarajan,Anna-Lena Schütz,Steven A. Johnsen,Stefan Bonn,Reinhardt Lührmann,Camin Dean,Andre Fischer,Andre Fischer +17 more
TL;DR: It is determined that aging and amyloid pathology are associated with inflammation and impaired synaptic function in the hippocampal CA1 region as the result of epigenetic-dependent alterations in gene expression, and histone deacetylase inhibitors should be further explored as a cost-effective therapeutic strategy against age-associated cognitive decline.
CDK9 directs H2B monoubiquitination and controls replication‐dependent histone mRNA 3′‐end processing
Judith Pirngruber,Andrei Shchebet,Lisa Schreiber,Efrat Shema,Neri Minsky,Rob D. Chapman,Dirk Eick,Yael Aylon,Moshe Oren,Steven A. Johnsen +9 more
TL;DR: It is shown that CDK9 activity is essential for maintaining global and gene‐associated levels of histone H2B monoubiquitination (H2Bub1) and help to maintain correct replication‐dependent histone messenger RNA (mRNA) 3′‐end processing.
Estrogen receptor isoform‐specific regulation of endogenous gene expression in human osteoblastic cell lines expressing either ERα or ERβ
David G. Monroe,Barbara Getz,Steven A. Johnsen,B. Lawrence Riggs,Sundeep Khosla,Thomas C. Spelsberg +5 more
TL;DR: The distinct patterns of E2‐dependent gene regulation in the U2OS cells by ERα and ERβ shown here suggest that during OB differentiation, a unique pattern of gene responses will occur, partially due to the differentiation state and the ER isoform present.
