TL;DR: Engineered synthetic GPCRs (PAGERs) enable programmable sensing and control of cell behavior in response to user-defined antigens, driving diverse cellular processes such as transgene expression, fluorescence, and G-protein activation, with broad utility in discovery and translational science.
Abstract: Synthetic receptors that mediate antigen-dependent cell responses are transforming therapeutics, drug discovery and basic research1,2. However, established technologies such as chimeric antigen receptors3 can only detect immobilized antigens, have limited output scope and lack built-in drug control3, 4, 5, 6–7. Here we engineer synthetic G-protein-coupled receptors (GPCRs) that are capable of driving a wide range of native or non-native cellular processes in response to a user-defined antigen. We achieve modular antigen gating by engineering and fusing a conditional auto-inhibitory domain onto GPCR scaffolds. Antigen binding to a fused nanobody relieves auto-inhibition and enables receptor activation by drug, thus generating programmable antigen-gated G-protein-coupled engineered receptors (PAGERs). We create PAGERs that are responsive to more than a dozen biologically and therapeutically important soluble and cell-surface antigens in a single step from corresponding nanobody binders. Different PAGER scaffolds allow antigen binding to drive transgene expression, real-time fluorescence or endogenous G-protein activation, enabling control of diverse cellular functions. We demonstrate multiple applications of PAGER, including induction of T cell migration along a soluble antigen gradient, control of macrophage differentiation, secretion of therapeutic antibodies and inhibition of neuronal activity in mouse brain slices. Owing to its modular design and generalizability, we expect PAGERs to have broad utility in discovery and translational science. Modular synthetic G-protein-coupled receptors with nanobody-based ligand-recognition domains can be designed and used to programme transgene expression, real-time fluorescence or endogenous G-protein activation in response to soluble or cell-surface ligands, enabling control of diverse cellular behaviours.
TL;DR: On September 17, 2024, thousands of pagers detonated in Beirut, Lebanon, injuring 2800 and killing 12, with most injuries in the face, eyes, or hands, prompting a hospital to assess disaster response plans and conduct regular exercises for future disasters.
Abstract: Abstract On September 17, 2024, at 15:30 local time, thousands of pagers used by members of a specific party group detonated across Lebanon. As a result of the explosions, 2800 were wounded and 12 lost their lives. Almost two-thirds of the injuries were in the face, eyes, or hands. The Lebanese American University Medical Center received 38 injured and admitted 36 patients, 13 of them to the Intensive care unit. A total of 33 patients needed surgeries. All medical and nursing staff were deployed. The health care workers faced major challenges that night: the severity of the injuries and the unprecedented types of injuries with the same pattern, and the urgent need for ophthalmology and orthopedics within the hospital and across the country. Learning from the Pager Explosion, each hospital should perform assessments of their disaster response plan, develop trainings, and conduct regular exercises in preparation for future disasters.

TL;DR: PAGER project successfully concluded in 2023, providing real-time space weather forecast and tools for satellite operators.
Abstract: The European Union's Horizon 2020 Prediction of Adverse effects of Geomagnetic storms and Energetic Radiation (PAGER) project was successfully concluded in 2023. This project provides real-time space weather forecast initiated from the solar observations as well as predictions of radiation in space and its effects on satellite infrastructure. Real-time predictions of particle radiation and cold plasma density allow for evaluation of surface charging and deep dielectric charging. The project provides a 1-2-day probabilistic and data assimilative forecast of ring current and radiation belt environments, which allows satellite operators to respond to predictions that present a significant threat. As a backbone of the project, we use the state-of-the-art codes that currently exist and adapt existing codes to perform ensemble simulations and uncertainty quantifications. Within PAGER, a number of innovative tools was obtained, including data assimilation and uncertainty quantification, new models of near-Earth electromagnetic wave environment, ensemble predictions of solar wind parameters at L1, and data-driven forecast of the geomangetic Kp index and plasma density. In this presentation, we show the overview of the outcomes and the products obtained within the project. The developed codes may be used in the future for realistic modelling of extreme space weather events.

TL;DR: This research examines the history, drivers, and perspectives of Financing for Development (FfD) negotiations at the United Nations, focusing on Brazil's engagement, amidst a paradigm shift in international cooperation, highlighting challenges, trends, and emerging areas of convergence.

TL;DR: Researchers introduce PAGER, a novel genotype encoding strategy that accurately models deviations from additivity in complex trait association studies, improving model precision and reducing computational costs, and identifies novel QTLs in real-world data.
Abstract: The additive model of inheritance assumes that heterozygotes (Aa) are exactly intermediate in respect to homozygotes (AA and aa). While this model is commonly used in single-locus genetic association studies, significant deviations from additivity are well-documented and contribute to phenotypic variance across many traits and systems. This assumption can introduce type I and type II errors by overestimating or underestimating the effects of variants that deviate from additivity. Alternative genotype encoding strategies have been explored to account for different inheritance patterns, but they often incur significant computational or methodological costs. To address these challenges, we introduce PAGER (Phenotype Adjusted Genotype Encoding and Ranking), an efficient pre-processing method that encodes each genetic variant based on normalized mean phenotypic differences between diallelic genotype classes (AA, Aa, and aa). This approach more accurately reflects each variant's true inheritance model, improving model precision while minimizing the costs associated with alternative encoding strategies. Through extensive benchmarking on SNPs simulated with both binary and continuous phenotypes, we demonstrate that PAGER accurately represents various inheritance patterns (including additive, dominant, recessive, and heterosis), achieves levels of statistical power that meet or exceed other encoding strategies, and attains computation speeds up to 55 times faster than a similar method, EDGE. We also apply PAGER to publicly available real-world data and identify a novel, relevant putative QTL associated with body mass index in rats (Rattus norvegicus) that is not detected with the additive model. Overall, we show that PAGER is an efficient genotype encoding approach that can uncover sources of missing heritability and reveal novel insights in the study of complex traits while incurring minimal costs.

Abstract: Effective communication is essential in the healthcare industry, as it can significantly improve workflow and positively impact patient care and healthcare professionals' (HCPs) working conditions. With the introduction of digital technologies, healthcare systems have been able to upgrade communication channels and streamline communication processes. Among these technologies, the pager system has been widely embraced by hospitals worldwide as a means to improve communication among HCPs. With progress in science and technology, new communication platforms have been developed and are now widely used in many sectors. Poor communication in hospitals affects four aspects of hospital work: efficiency of resource utilisation, effectiveness of core operations, quality of HCP’s work life and quality of service provided in the hospital. This project examines these issues in the UK National Health Service by comparing the traditional pager system with widely available multi-functional mobile phone communications apps such as WhatsApp and bespoke apps such as Alertive and Medic Bleep. A survey of HCPs and some in-depth interviews were conducted to examine the lived experience of working with pagers and apps. The results show that HCPs have reported that using modern, multi-functional digital communication apps can significantly enhance workflow efficiency within the hospital. This, in turn, can positively impact various aspects of hospital operations, such as resource utilisation, patient care, and staff work-life balance. The project recommends that healthcare managers consider investing in communication apps that offer multiple functionalities to replace traditional pagers. However, it is essential to acknowledge that implementing new technology may present challenges in terms of change management.
