Airborne persistent toxic substances are associated with health impacts resulting from air pollution, for example, dioxins, dioxin-like polychlorinated biphenyls, and certain polycyclic aromatic hydrocarbons (PAHs), which activate aryl hydrocarbon receptors (AhR) and thereby produce adverse outcomes. Thus, a bioassay for evaluating AhR activation is required for risk assessment of ambient-air samples, and for this purpose, we developed a new and sensitive recombinant mouse hepatoma cell line, CBG2.8D, in which a novel luciferase-reporter plasmid containing two copies of a newly designed dioxin-responsive domain and a minimal promoter derived from a native gene were integrated. The minimal detection limit for 2,3,7,8-tetrachlorodibenzo- p-dioxin with this assay system was 0.1 pM. We used CBG2.8D to determine dioxin levels in 45 ambient-air samples collected in Beijing. The measured bioanalytical equivalent (BEQ) values were closely correlated with the toxic equivalent values obtained from chemical analysis. In haze ambient-air samples, the total activation of aryl hydrocarbon receptors (TAA) was considerably higher than the BEQ of dioxin-rich fractions, according to the results of the cell-based bioassay. Notably, the haze samples contained abundant amounts of PAHs, whose relative toxicity equivalent was correlated with the TAA; this finding suggests that PAHs critically contribute to the AhR-related biological impacts of haze ambient-air samples.

Development and Application of a Novel Bioassay System for Dioxin Determination and Aryl Hydrocarbon Receptor Activation Evaluation in Ambient-Air Samples.

Ambient air was sampled and analyzed around a municipal solid waste incinerator (MSWI) in Beijing from April 2014 to January 2015 to investigate the concentrations, profiles and seasonal variations of PCDD/Fs in the region using HRGC-HRMS technique. The mass concentrations and TEQ of 2,3,7,8-substituted PCDD/Fs in the air samples ranged from 8.9 to 140 pg·m-3 and from 0.11 to 1.8 pg·m-3, respectively. The concentration values at 4 sampling sites in haze day in autumn and all 7 sampling sites in winter were higher than the ambient air standard of 0.6 pg·m-3 for dioxins regulated in Japan. 1,2,3,4,6,7,8-HpCDF and OCDD dominated PCDD/Fs in all the samples for all four seasons with average contribution fractions of 20.5% and 14.0%, respectively, while 2,3,4,7,8-PeCDF was the dominant congener contributing to TEQ (43.3%). The spatial distribution basically exhibited a trend that the concentrations at all sites were comparable and not related to the distances from the source. Seasonal variation showed obviously higher concentration in winter than the other three seasons, which may attribute to the high concentration of ambient particulate matter due to domestic heating and worse atmospheric dispersion that occurred in winter. The homologue and congener profiles of PCDD/Fs in the air samples differed from those of the flue gas emission from the MSWI, consistent with the principle component analysis results. Dioxin inhalation exposure dose estimation showed that the dioxin inhalation exposure risk of residents living in the studied area was at a relatively safe level[0.060-0.224 pg·(kg·d)-1]. However, the dioxin inhalation exposure risk in heavily polluted seasons still needs great concerns.

[Pollution Characteristics of PCDD/Fs in Ambient Air and Exposure Risk Assessment Around a Municipal Solid Waste Incinerator in Beijing].

Air samples were collected from six locations during the periods of February, 2011-March, 2012 in Beijing city. Samples were analyzed using HRGC-HRMS according to HJ 77. 2-2008. The concentrations, congener profiles, seasonal variation, spatial distribution, and sources identified were investigated. The mass concentrations of 2,3,7,8-substituted PCDD/Fs is 1750 to 10380 fg/m(3) with an average of 3670 fg/m(3). The highest concentration was observed at site DS in February 2011 and the lowest was at site NS in July 2011. The TEQ concentrations of PCDD/Fs ranged from 35.0 to 751 fg I-TEQ/m(3), with an average of 251 fg I-TEQ/m(3). The dominant contributor to total TEQ value is 2,3,7,8-PeCDFs, with an average contribution of 40%. Low-chlorinated PCDDs were observed in all samples in winter. The concentration and profiles exhibit obvious seasonal variation. The PCDD/Fs at six sampling sites did not show obvious spatial variation. Seasonal variation of PCDD/F concentrations indicated that the emission sources may be different. Fossil fuel combustion and automobiles may be the main dioxins emission sources during the heating period, whereas, in non-heating period, the automobiles alone are the main emission sources.
