A membrane-penetrating cation, dodecyltriphenylphosphonium (C12TPP), facilitates the recycling of fatty acids in the artificial lipid membrane and mitochondria. C12TPP can dissipate mitochondrial membrane potential and may affect total energy expenditure and body weight in animals and humans. We investigated the metabolic effects of C12TPP in isolated brown-fat mitochondria, brown adipocyte cultures and mice in vivo. Experimental approaches included the measurement of oxygen consumption, carbon dioxide production, western blotting, magnetic resonance imaging and bomb calorimetry. In mice, C12TPP (50 μmol per (day•kg body weight)) in the drinking water significantly reduced body weight (12%, P<0.001) and body fat mass (24%, P<0.001) during the first 7 days of treatment. C12TPP did not affect water palatability and intake or the energy and lipid content in feces. The addition of C12TPP to isolated brown-fat mitochondria resulted in increased oxygen consumption. Three hours of pretreatment with C12TPP also increased oligomycin-insensitive oxygen consumption in brown adipocyte cultures (P<0.01). The effects of C12TPP on mitochondria, cells and mice were independent of uncoupling protein 1 (UCP1). However, C12TPP treatment increased the mitochondrial protein levels in the brown adipose tissue of both wild-type and UCP1-knockout mice. Pair-feeding revealed that one-third of the body weight loss in C12TPP-treated mice was due to reduced food intake. C12TPP treatment elevated the resting metabolic rate (RMR) by up to 18% (P<0.05) compared with pair-fed animals. C12TPP reduced the respiratory exchange ratio, indicating enhanced fatty acid oxidation in mice. C12TPP combats diet-induced obesity by reducing food intake, increasing the RMR and enhancing fatty acid oxidation.

Mitochondria-targeted dodecyltriphenylphosphonium (C12TPP) combats high-fat-diet-induced obesity in mice.

The present study aimed to examine the effects of a prior session of power training on blood pressure values during tasks that mimic the activities of daily living in hypertensive and normotensive older women. A randomized crossover experimental design was used for this study. Eleven older women (age, 66.1±1.2 years; body mass index, 26.7±4.8 kg/m2; systolic blood pressure, 133.9±23.4 mmHg) were recruited to participate. Volunteers were randomly underwent to a single session of physical exercise equalized by training volume, characterized by 3 sets of 8-10 repetitions in 8 different exercises. However, Power training session was performed at a higher intensity (moderate) than Sham session (rest/very, very easy ). Immediately after the end of the exercise session, subjects were assigned to perform five tasks that mimic the activities of daily living. Hemodynamic parameters were recorded before and immediately after the session of physical exercise, as well as in the end of each of the tasks. Results demonstrated significant elevations in systolic blood pressure and pulse pressure values during the performance of the tasks after the Sham session. However, values were not altered in the power training session. Data of the present study indicate that an acute session of power training can maintain blood pressure values lower during the subsequent performance of tasks that mimic activities of daily living in older women.

Berardinelli-Seip Congenital Lipodystrophy (BSCL) is a rare autosomal recessive disease characterized by the almost complete absence of adipose tissue. Due to a strong founder effect that resulted in a higher prevalence of BSCL in Rio Grande do Norte (RN), a state in northeastern Brazil, it has been essential that health professionals develop knowledge about this disease. Nurses are often the first point of contact with patients during health care assistance. The purpose of this study was to investigate the knowledge of these professionals about BSCL in two main hospitals in RN state. A questionnaire was applied to 199 nurses working in the Hospital Regional Mariano Coelho-HRMC (Regional Hospital Mariano Coelho), in Currais Novos-RN, and in the Hospital Universitario Onofre Lopes-HUOL (University Hospital Onofre Lopes), in Natal-RN. This study showed that most nursing professionals do not know about the disease, although they have already received patients with BSCL in those hospitals. The nurses from HRMC and HUOL lacked knowledge of BSCL and the healthcare of these patients requires immediate improvement. Significant efforts are required to close the gap between current and needed practice patterns.

Objective: To evaluate the interference of supervised resistance and aerobic training in glycemic levels during and after 36 sessions, belonging to a training period of three months in patients with type 2 diabetes mellitus (T2DM), as well as the comparison of both exercise training. Methods: This is a experimental study with twelve T2DM patients, all sedentary and attended by the Specialized Care Center of Vicosa-MG, randomized into aerobic (n = 6.54 ± 5 years) and resisted (n = 6.58 ± 9 years) groups. Capillary glycemia was collected before (M1), during (M2) and after (M3) each exercise session for 12 weeks with a weekly frequency of three days. It was used ANOVA for repeated measures followed by Tukey and Student’s test. Results: A mean drop in capillary glycemia was observed between M1 and M2 (p = 0.001), M2 and M3 (p = 0.001) and M1 and M3 (p = 0.001) in both groups. Conclusion: It is concluded that aerobic and resistance exercise training caused a decreased in the blood glucose during exercise sessions. The findings showed that both modes of exercise are important in the hyperglycemic treatment of T2DM patients.

The literature has shown the efficiency of exercise in the control of type 2 diabetes(T2D), being suggested as one of the best kinds of non-pharmacological treatments for its population. Thus, the scientific production related to this phenomenon has growing exponentially. However, despite its advances, still there is a lack of studies that have carried out a review on the acute effects of physical exercise on metabolic and hemodynamic markers and possible control mechanisms of these indicators in individuals with T2 D, not to mention that in a related way, these themes have been very little studied today. Therefore, the aim of this study was to organize and analyze the current scientific production about the acute effects of physical exercise on metabolic and hemodynamic markers and possible control mechanisms of these indicators in T2 D individuals. For such, a research with the following keywords was performed:-exercise; diabetes and post-exercise hypotension; diabetes and excess post-exercise oxygen consumption; diabetes and acute effects in PUBMED, SCIELO and HIGHWIRE databases. From the analyzed studies, it is possible to conclude that, a single exercise session can promote an increase in the bioavailability of nitric oxide and elicit decreases in postexercise blood pressure. Furthermore, the metabolic stress from physical exercise can increase the oxidation of carbohydrate during the exercise and keep it, in high levels, the post exercise consumption of O2, this phenomenon increases the rate of fat oxidation during recovery periods after exercise, improves glucose tolerance and insulin sensitivity and reduces glycemia between 2-72 h, which seems to be dependent on the exercise intensity and duration of the effort.
