In vitro induced tetraploid of Dendranthema nankingense (Nakai) Tzvel. shows an improved level of abiotic stress tolerance

Chrysanthemum is globally the second most important ornamental in terms of socioeconomic importance. Even though the vast range of flower colors, shapes and forms were initially created using conventional and mutation breeding, transgenic strategies are now more frequently used with Agrobacterium-mediated transformation being the most popular form of introducing foreign genes into chrysanthemums. Even so, transformation efficiency remains dependent on cultivar and regeneration procedure. Transgenic molecular breeding has seen the introduction of important traits such as novel flower color and form and plant architecture, prolonged cut-flower vase-life, resistance to biotic stresses such as viruses/viroids, pathogens and insects. However, chimerism and transgene silencing continue to be limiting factors. Transgenic strategies, despite opening up new avenues for creating new cultivars with improved agronomic and horticultural traits, may be limited due to the risk of transgenic pollen escaping into the wild.

Chrysanthemum Biotechnology: Quo vadis?

A major constraint affecting the quality and productivity of chrysanthemum is the unusual period of low temperature occurring during early spring, late autumn, and winter. Yet, there has been no systematic investigation on the genes underlying the response to low temperature in chrysanthemum. Herein, we used RNA-Seq platform to characterize the transcriptomic response to low temperature by comparing different transcriptome of Chrysanthemum nankingense plants and subjecting them to a period of sub-zero temperature, with or without a prior low temperature acclimation. Six separate RNA-Seq libraries were generated from the RNA samples of leaves and stems from six different temperature treatments, including one cold acclimation (CA), two freezing treatments without prior CA, two freezing treatments with prior CA and the control. At least seven million clean reads were obtained from each library. Over 77% of the reads could be mapped to sets of C. nankingense unigenes established previously. The differentially transcribed genes (DTGs) were identified as low temperature sensing and signalling genes, transcription factors, functional proteins associated with the abiotic response, and low temperature-responsive genes involved in post-transcriptional regulation. The differential transcription of 15 DTGs was validated using quantitative RT-PCR. The large number of DTGs identified in this study, confirmed the complexity of the regulatory machinery involved in the processes of low temperature acclimation and low temperature/freezing tolerance.

Ornamentals come in a variety of shapes, sizes, and colors to suit a wide range of climates, landscapes, and gardening needs. Compared to demand, a shortage of plant materials and diversity force the search for solutions for their constant acquisition and improvement to increase their commercial value, respectively. In vitro cultures are a suitable solution to meet expectations using callus culture, somatic embryogenesis, protoplast culture, and the organogenesis of protocorm-like bodies; many of these techniques are commercially practiced. Factors such as culture media, explants, carbohydrates, plant growth regulators, and light are associated with the success of in vitro propagation. Techniques, especially embryo rescue and somatic hybridization, are widely used to improve ornamentals. The development of synthetic seed allows season-independent seed production and preservation in the long term. Despite the advantages of propagation and the improvement of ornamentals, many barriers still need to be resolved. In contrast to propagation and crop developmental studies, there is also a high scope for molecular studies, especially epigenetic changes caused by plant tissue culture of ornamentals. In this review, we have accumulated and discussed an overall update on cultivation factors, propagation techniques in ornamental plant tissue culture, in vitro plant improvement techniques, and future perspectives.

Tissue Culture in Ornamentals: Cultivation Factors, Propagation Techniques, and Its Application

The interspecific cross between Chrysanthemum × grandiflorum (Ramat.) Tzvel. ‘rm20-12’ (R, 2n = 54) and C. makinoi Matsum., and Nakai (M, 2n = 18) was achieved using embryo rescue, and a single backcross progeny using C. × grandiflorum ‘rm20-12’ as paternal parent was obtained. The morphology of the two independent F1 hybrids (MR1 and MR2) differed from that of both parents. MR1 had a larger inflorescence diameter along with narrow leaves and a reduced number of ray and tubular florets. MR2 was shorter and its inflorescence developed fewer tubular florets than either M or R. The BC1F1 hybrid was similar to its maternal plant MR2 in terms of leaf length and width, inflorescence diameter and the number of ray florets, while it produced fewer tubular florets than either MR2 or R. The flower color in both F1 hybrids was lavender, while the BC1F1 plant bore purple flowers. The aphid resistance and heat tolerance of MR1, MR2 and the BC1F1 hybrid were both significantly superior to that of C. × grandiflorum ‘rm20-12’. Interspecific hybridization followed by backcrossing shows clear potential for cultivar improvement in chrysanthemum.

Creating novel chrysanthemum germplasm via interspecific hybridization and backcrossing

Ovary rescue was employed to create six interspecific hybrids from the cross between Dendranthema morifolium (Ramat.) Kitamura ‘rm20-12’ (2n = 54) and its wild diploid relative D. nankingense (Nakai) Tzvel. (2n = 18). The morphology of the hybrids differed from that of either parent. The leaf length and width of all three D. morifolium × D. nankingense hybrids exceeded that of the parents, as did the plant height of two and the inflorescence diameter of another of the hybrids. One of the reciprocal hybrids was heterotic for leaf length and width. All the hybrids bore yellow flowers. The cold tolerance of five hybrids was significantly superior to that of their D. morifolium parent. Interspecific hybridization clearly provides an effective means of cultivar improvement in chrysanthemum.
