Fluoroquinolones of the third and fourth generation posses wide bactericidal activity. Monitoring concentrations of antibacterial agents provides effective therapy and prevents the increase of bacterial resistance to antibiotics. The pharmacodynamic parameters that best describe fluoroquinalone activity are AUC/MIC and C
 max/MIC. Determining the level of this type of drug is essential to reach the effective concentration that inhibits the growth of bacteria. Determining the pharmaceutical formulation confirms the purity of a substance. Many methods have been developed to determine the level of these substances. They involve mainly the following analytical techniques: chromatography, capillary electrophoresis, and spectroscopy. The separation techniques were combined with different measuring devices, such as ultraviolet (UV), fluorescence detector (FLD), diode array detector (DAD), and mass spectrometry (MS). The analytical procedures require proper sample pre-conditioning such as protein precipitation, extraction techniques, filtration, or dilution. This paper reviews the reported analytical methods for the determining representatives of the third and fourth generation of fluoroquinolones. Attention was paid to pre-conditioning of the samples and the applied mobile phase. This report might be helpful in the selection of the proper procedure in determining the abovementioned drugs in different matrices.

Analytical Methods for Determining Third and Fourth Generation Fluoroquinolones: A Review.

A fast method for simultaneous determination of amantadine, rimantadine, and chlorpheniramine in seven animal derived samples including pork, chicken, duck, pig liver, chicken liver, pig kidney, and egg was developed using liquid chromatography-tandem mass spectrometry, and employed a new multifunctional syringe filter that makes the cleanup procedure simple and rapid based on the QuEChERS (quick, easy, cheap, effective, rugged and safe) method. The method was validated using amantadine-d15, rimantadine-d4 and chlorpheniramine-d6 as internal standards for three analytes. Good linearities (R2 > 0.9938) were obtained over the concentration range from 2 μg L−1 to 200 μg L−1 for amantadine and rimantadine, and from 0.2 μg L−1 to 20 μg L−1 for chlorpheniramine. The precision was evaluated by intra- and inter-day assays and the relative standard deviations were all within 9.85%. Mean recoveries ranged from 89.9% to 105%. The limits of detection and quantification were 0.5 and 1.0 μg kg−1 for both amantadine and rimantadine, and 0.05 and 0.1 μg kg−1 for chlorpheniramine, respectively. The application of the developed method in real samples showed that amantadine and chlorpheniramine were respectively detected with percentages of 3.7% and 0.3% in all tested samples.

The prerequisites for forensic confirmatory analysis by LC/MS/MS with respect to European Union guidelines are chromatographic separation, a minimum number of two MS/MS transitions to obtain the required identification points and predefined thresholds for the variability of the relative intensities of the MS/MS transitions (MRM transitions) in samples and reference standards. In the present study, a fast, sensitive and robust method to quantify tramadol, chlorpheniramine, dextromethorphan and their major metabolites, O-desmethyltramadol, dsmethyl-chlorpheniramine and dextrophan, respectively, in human plasma using ibuprofen as internal standard (IS) is described. The analytes and the IS were extracted from plasma by a liquid–liquid extraction method using ethyl acetate–diethyl-ether (1:1). Extracted samples were analyzed by ultra-high-performance liquid chromatography coupled to electrospray ionization tandem mass spectrometry (UHPLC-ESI-MS/MS). Chromatographic separation was performed by pumping the mobile phase containing acetonitrile, water and formic acid (89.2:11.7:0.1) for 2.0 min at a flow rate of 0.25 μL/min into a Hypersil-Gold C18 column, 20 × 2.0 mm (1.9 µm) from Thermoscientific, New York, USA. The calibration curve was linear for the six analytes. The intraday precision (RSD) and accuracy (RE) of the method were 3–9.8 and −1.7–4.5%, respectively. The analytical procedure herein described was used to assess the pharmacokinetics of the analytes in 24 healthy volunteers after a single oral dose containing 50 mg of tramadol hydrochloride, 3 mg chlorpheniramine maleate and 15 mg of dextromethorphan hydrobromide. Development and validation of a sensitive UHPLC‐MS/MS method for the simultaneous analysis of tramadol, dextromethorphan chlorpheniramine and their major metabolites in human plasma in forensic context: application to pharmacokinetics

Dextromethorphan (DXM) is an antitussive drug found in commonly used nonprescription cold and cough medications. At low doses, DXM is a safe drug that does not produce adverse reactions. However, abuse of DXM has been reported among adolescents and young adults using the drug at higher doses. DXM is not a scheduled drug in the USA, and the primary reason for its abuse is the ease of availability. DXM is available to purchase in the form of over-the-counter cough medications, such as Robitussin(®) and Coricidin(®), or it can be purchased over the Internet in the form of a powder. In this research work, we developed an LC-MS-MS method that can quantify DXM and dextrorphan (DXO) in oral fluid in a high-throughput toxicology laboratory setting. The developed method was validated according to the Scientific Working Group for Forensic Toxicology guidelines. The linear dynamic range was 5-100 ng/mL with a lowest limit of quantitation (LLOQ) of 5.0 ng/mL for DXM and DXO. Overall, the results of the accuracy and the precision values were within the acceptance criteria for both drugs. In addition, selectivity, matrix effect and recovery were calculated for the LC-MS-MS method. Authentic samples (n = 59) were tested to evaluate the applicability of the method. Thirty samples were found to be positive for DXM and DXO and two samples were found to be positive for DXM only.

Determination of sitafloxacin in human plasma by liquid chromatography-tandem mass spectrometry method: application to a pharmacokinetic study.

A sensitive and accurate HPLC-MS/MS method was developed for the simultaneous determination of dextromethorphan, dextrorphan and chlorphenamine in human plasma. Three analytes were extracted from plasma by liquid–liquid extraction using ethyl acetate and separated on a Kromasil 60-5CN column (3 µm, 2.1 × 150 mm) with mobile phase of acetonitrile–water (containing 0.1% formic acid; 50:50, v/v) at a flow rate of 0.2 mL/min. Quantification was performed on a triple quadrupole tandem mass spectrometer in multiple reaction monitoring mode using positive electrospray ionization. The calibration curve was linear over the range of 0.01–5 ng/mL for dextromethorphan, 0.02–5 ng/mL for dextrorphan and 0.025–20 ng/mL for chlorphenamine. The lower limits of quantification for dextromethorphan, dextrorphan and chlorphenamine were 0.01, 0.02 and 0.025 ng/mL, respectively. The intra- and inter-day precisions were within 11% and accuracies were in the range of 92.9–102.5%. All analytes were proved to be stable during sample storage, preparation and analytic procedures. This method was first applied to the pharmacokinetic study in healthy Chinese volunteers after a single oral dose of the formulation containing dextromethorphan hydrobromide (18 mg) and chlorpheniramine malaeate (8 mg).
