Highly efficient CRISPR/Cas9-mediated transgene knockin at the H11 locus in pigs.
Jinxue Ruan,Li Hegang,Kui Xu,Tianwen Wu,Wei Jingliang,Rong Zhou,Zhiguo Liu,Yulian Mu,Shulin Yang,Hongsheng Ouyang,Ruby Yanru Chen-Tsai,Kui Li +11 more
TL;DR: This work describes for the first time the generation of a site-specific knockin pig model using a combination of CRISPR/Cas9 and somatic cell nuclear transfer and reports a new genomic “safe harbor” locus, named pH11, which enables stable and robust transgene expression.
CD163 and pAPN double-knockout pigs are resistant to PRRSV and TGEV and exhibit decreased susceptibility to PDCoV while maintaining normal production performance.
Kui Xu,Yanrong Zhou,Yulian Mu,Zhiguo Liu,Shaohua Hou,Yujian Xiong,Liurong Fang,Changli Ge,Wei Yinghui,Zhang Xiuling,Changjiang Xu,Jingjing Che,Ziyao Fan,Guangming Xiang,Jiankang Guo,Haitao Shang,Hua Li,Shaobo Xiao,Li Julang,Kui Li +19 more
TL;DR: Generation of double-gene-knockout (DKO) pigs harboring edited knockout alleles for known receptor proteins CD163 and pAPN are reported and it is shown that DKO pigs are completely resistant to genotype 2 PRRSV and TGEV.

Mitochondrial base editor DdCBE causes substantial DNA off-target editing in nuclear genome of embryos
Ying-Ze Wei,Zhifang Li,Kui Xu,Hu Feng,Long Xie,Di Li,Zhenrui Zuo,Meiling Zhang,Chunlong Xu,Hui Yang,Erwei Zuo +10 more
TL;DR: DdCBE has the potential to model mitochondrial disease mutations, correct pathogenic variants, and expand the knowledge of mitochondrial biology, but the off-target effect remained to be comprehensively investigated by additional research for their systematic effect on mtDNA as well as nuclear genome.
Human cleaving embryos enable efficient mitochondrial base-editing with DdCBE
Ying-Ze Wei,Chunlong Xu,Hu Feng,Kui Xu,Zhifang Li,Jing Hu,Ling Zhou,Yu Wei,Zhenrui Zuo,Erwei Zuo,Wen Li,Hui Yang,Meiling Zhang +12 more
TL;DR: DdCBE’s ability for base editing of mtDNA in human embryos is tested and it is found that cleaving embryos enable robust base conversion in nuclear DNA, similar to the recent finding in HEK293T cells.
Integrative Proteomic and Phosphoproteomic Profiling of Testis from Wip1 Phosphatase-Knockout Mice: Insights into Mechanisms of Reduced Fertility.
Wei Yinghui,Qian Gao,Pengxia Niu,Kui Xu,Yiqing Qiu,Yanqing Hu,Shasha Liu,Xue Zhang,Miaoying Yu,Zhiguo Liu,Bingyuan Wang,Yulian Mu,Kui Li +12 more
TL;DR: The mechanism that proinflammatory cytokines may impair the blood-testis barrier dynamics by decreasing the expression of junction-associated proteins in Wip1 null testes, leading to subfertility and spermatogenesis defects is established.
