mRNA therapeutics deliver a hopeful message
Zifu Zhong,Séan Mc Cafferty,Francis Combes,Hanne Huysmans,Joyca De Temmerman,Arlieke Gitsels,Daisy Vanrompay,João Paulo Portela Catani,Niek N. Sanders +8 more
TL;DR: The different synthetic mRNA platforms, their production and purification, strategies to tackle the inherent innate immunogenicity of synthetic mRNA, the recent progress in the development of nanoformulations for the in vivo delivery of mRNA, as well as the efficacy and safety of mRNA therapeutics in recent clinical trials are described.

Corticosteroids and cellulose purification improve, respectively, the in vivo translation and vaccination efficacy of sa-mRNAs.
Zifu Zhong,Séan McCafferty,Lisa Opsomer,Haixiu Wang,Hanne Huysmans,Joyca De Temmerman,Stefan Lienenklaus,João Paulo Portela Catani,Francis Combes,Niek N. Sanders +9 more
TL;DR: In this article, the effect of several innate immune inhibitors and a novel cellulose-based mRNA purification approach on the type I interferon (IFN) response and the translation and vaccination efficacy of a sa-mRNA vaccine against Zika virus was investigated.

In Vivo Validation of a Reversible Small Molecule-Based Switch for Synthetic Self-Amplifying mRNA Regulation.
Séan Mc Cafferty,Joyca De Temmerman,Tasuku Kitada,Jacob R. Becraft,Ron Weiss,Darrell J. Irvine,Mathias Devreese,Siegrid De Baere,Francis Combes,Niek N. Sanders +9 more
TL;DR: The in vivo utility of a synthetic self-amplifying mRNA (RNA replicon) whose expression can be turned off using a genetic switch that responds to oral administration of trimethoprim (TMP), an FDA-approved small-molecule drug is validated.

Mononuclear but Not Polymorphonuclear Phagocyte Depletion Increases Circulation Times and Improves Mammary Tumor-Homing Efficiency of Donor Bone Marrow-Derived Monocytes.
Francis Combes,Alexandros Marios Sofias,Séan Mc Cafferty,Hanne Huysmans,Joyca De Temmerman,Sjoerd Hak,Evelyne Meyer,Niek N. Sanders +7 more
TL;DR: Using immune cell depleting agents and macroscopic in vivo fluorescence imaging, it is demonstrated that removal of endogenous monocytes and macrophages leads to an increased tumor accumulation of exogenously administered BMDMs and that clodronate-mediated depletion drastically increases the circulation time of the exogenous BMDs.

The impact of hepatocyte-specific deletion of hypoxia-inducible factors on the development of polymicrobial sepsis with focus on GR and PPARα function
Tineke Vanderhaeghen,Steven Timmermans,Melanie Eggermont,Deepika Watts,Jolien Vandewalle,Charlotte Wallaeys,Louise Nuyttens,Joyca De Temmerman,Tino Hochepied,Sylviane Dewaele,Joke Vanden Berghe,Niek N. Sanders,Ben Wielockx,Rudi Beyaert,Claude Libert +14 more
TL;DR: In this paper , the authors investigated the hypothesis that hypoxia and/or activation of Hypoxia inducible Factor (HIFs) might play a role in these issues with hepatic peroxisome proliferator-activated receptor alpha (PPARα) and glucocorticoid receptor (GR) and concluded that HIF1α and HIF2α are activated in hepatocytes in sepsis, but their contribution to the mechanisms leading to lethality are minimal.
