Physiological blood-brain transport is impaired with age by a shift in transcytosis.
Andrew C. Yang,Marc Y. Stevens,Michelle B. Chen,Davis P. Lee,Daniel Stähli,David Gate,Kévin Contrepois,Winnie Chen,Tal Iram,Lichao Zhang,Ryan T. Vest,Aisling Chaney,Benoit Lehallier,Niclas Olsson,Haley du Bois,Ryan Hsieh,Haley C. Cropper,Daniela Berdnik,Lulin Li,Elizabeth Y. Wang,Gavin M. Traber,Carolyn R. Bertozzi,Carolyn R. Bertozzi,Jian Luo,Jian Luo,Michael Snyder,Joshua E. Elias,Stephen R. Quake,Michelle L. James,Tony Wyss-Coray +29 more
TL;DR: The extent of physiological protein transcytosis to the healthy brain is revealed, a mechanism of widespread BBB dysfunction with age and a strategy for enhanced drug delivery is revealed.

11C-DPA-713 Versus 18F-GE-180: A Preclinical Comparison of Translocator Protein 18 kDa PET Tracers to Visualize Acute and Chronic Neuroinflammation in a Mouse Model of Ischemic Stroke
Aisling Chaney,Haley C. Cropper,Emily M. Johnson,Kendra J. Lechtenberg,Todd C. Peterson,Marc Y. Stevens,Marion S. Buckwalter,Michelle L. James +7 more
TL;DR: 11C-DPA-713 PET reflects the extent of microglial activation in infarcted distal middle cerebral artery occlusion mouse brain tissue more accurately than 18F-GE-180 and appears to be slightly more sensitive and warrant further investigation in both preclinical and clinical settings.

Longitudinal translocator protein-18 kDa-positron emission tomography imaging of peripheral and central myeloid cells in a mouse model of complex regional pain syndrome.
Haley C. Cropper,Emily M. Johnson,Elena S. Haight,Stephanie A. Cordonnier,Aisling Chaney,Thomas E. Forman,Anjali Biswal,Marc Y. Stevens,Michelle L. James,Vivianne L. Tawfik +9 more
TL;DR: Longitudinal positron emission tomography of translocator protein-18 kDa revealed early central, and persistent peripheral, myeloid activation in a mouse tibial fracture model of complex regional pain syndrome.

PET Imaging of Neuroinflammation Using [11C]DPA-713 in a Mouse Model of Ischemic Stroke.
TL;DR: A method to accurately quantify neuroinflammation using [11C]N,N-Diethyl-2-[2-methoxyphenyl)-5,7-dimethylpyrazolo[1,5-a]pyrimidin-3-yl]acetamide, a promising second generation TSPO-PET radiotracer, in distal middle cerebral artery occlusion (dMCAO) compared to sham-operated mice is presented.
