Disruption of a Large Intergenic Noncoding RNA in Subjects with Neurodevelopmental Disabilities
Michael E. Talkowski,Michael E. Talkowski,Gilles Maussion,Liam Crapper,Jill A. Rosenfeld,Ian Blumenthal,Carrie Hanscom,Colby Chiang,Amelia M. Lindgren,Shahrin Pereira,Douglas M. Ruderfer,Alpha B. Diallo,Juan Pablo Lopez,Gustavo Turecki,Elizabeth Suchi Chen,Carolina Oliveira Gigek,David J. Harris,Va Lip,Yu An,Yu An,Marta Biagioli,Marcy E. MacDonald,Marcy E. MacDonald,Michael F. Lin,Michael F. Natowicz,Yiping Shen,Yiping Shen,Yiping Shen,Cynthia C. Morton,Cynthia C. Morton,James F. Gusella,James F. Gusella,Carl Ernst +42 more
TL;DR: DNA capture followed by next-generation sequencing of the translocation breakpoints revealed disruption of a single noncoding gene on chromosome 2, LINC00299, whose RNA product is expressed in all tissues measured, but most abundantly in brain.
A molecular model for neurodevelopmental disorders
Carolina Oliveira Gigek,Elizabeth Suchi Chen,Elizabeth Suchi Chen,Vanessa Kiyomi Ota,Vanessa Kiyomi Ota,Gilles Maussion,Huashan Peng,Kathryn Vaillancourt,Alpha B. Diallo,Juan Pablo Lopez,Liam Crapper,Cristina Vasuta,Gang G. Chen,Carl Ernst +13 more
TL;DR: It is shown that reduced dosage of either gene leads to significant overlap of gene-expression patterns, microRNA patterns and DNA methylation states with control NSCs in a differentiating state, suggesting that a unifying feature of 2q23.1 and 2q33.1 deletion syndrome may be a lack of regulation between proliferation and differentiation in NSCS.
Highly penetrant alterations of a critical region including BDNF in human psychopathology and obesity

TL;DR: Hemizygosity of the BDNF region contributes to variable psychiatric phenotypes including anxiety, behavioral, and mood disorders as well as physical or cognitive impairment.

BisQC: an operational pipeline for multiplexed bisulfite sequencing
Gary G. Chen,Alpha B. Diallo,Raphaël Poujol,Corina Nagy,Alfredo Staffa,Kathryn Vaillancourt,Pierre-Eric Lutz,Vanessa Kiyomi Ota,Deborah C. Mash,Gustavo Turecki,Carl Ernst +10 more
TL;DR: BisQC, a step-by-step method for multiplexed bisulfite-converted DNA library construction, pooling, spike-in content, and bioinformatics, finds that decoupling amplification of bisulfITE converted (bis) DNA from the indexing reaction is an advantage, specifically in reducing total PCR cycle number and pre-selecting high quality bis-libraries.
Molecular Convergence of Neurodevelopmental Disorders
Elizabeth Suchi Chen,Carolina Oliveira Gigek,Jill A. Rosenfeld,Alpha B. Diallo,Gilles Maussion,Gary G. Chen,Kathryn Vaillancourt,Juan Pablo Lopez,Liam Crapper,Raphaël Poujol,Lisa G. Shaffer,Guillaume Bourque,Carl Ernst +12 more
TL;DR: The data suggest that 18q21 and 9q34 deletion syndromes show significant molecular convergence but distinct expression and methylation profiles, and common intersection points might highlight the most salient features of disease and provide avenues for similar treatments for NDDs caused by different genetic mutations.
