Gut Dysbiosis and Detection of “Live Gut Bacteria” in Blood of Japanese Patients With Type 2 Diabetes
Junko Sato,Akio Kanazawa,Fuki Ikeda,Tomoaki Yoshihara,Hiromasa Goto,Hiroko Abe,Koji Komiya,Minako Kawaguchi,Tomoaki Shimizu,Takeshi Ogihara,Yoshifumi Tamura,Yuko Sakurai,Risako Yamamoto,Tomoya Mita,Yoshio Fujitani,Hiroshi Fukuda,Koji Nomoto,Takuya Takahashi,Takashi Asahara,Takahisa Hirose,Satoru Nagata,Yuichiro Yamashiro,Hirotaka Watada +22 more
TL;DR: This is the first report of gut dysbiosis in Japanese patients with type 2 diabetes as assessed by RT-qPCR, and the high rate of gut bacteria in the circulation suggests translocation of bacteria from the gut to the bloodstream.
The diabetes-susceptible gene SLC30A8/ZnT8 regulates hepatic insulin clearance.
Motoyuki Tamaki,Yoshio Fujitani,Akemi Hara,Toyoyoshi Uchida,Yoshifumi Tamura,Kageumi Takeno,Minako Kawaguchi,Takahiro Watanabe,Takeshi Ogihara,Ayako Fukunaka,Tomoaki Shimizu,Tomoya Mita,Akio Kanazawa,Mica Ohara Imaizumi,Takaya Abe,Hiroshi Kiyonari,Shintaro Hojyo,Toshiyuki Fukada,Takeshi Kawauchi,Shinya Nagamatsu,Toshio Hirano,Ryuzo Kawamori,Hirotaka Watada +22 more
TL;DR: It is demonstrated that zinc secreted in concert with insulin suppressed hepatic insulin clearance by inhibiting clathrin-dependent insulin endocytosis and that genetic dysregulation of this system may play a role in the pathogenesis of type 2 diabetes.
Genetic variations in the gene encoding ELMO1 are associated with susceptibility to diabetic nephropathy.
Atsuyuki Shimazaki,Yoshihiro Kawamura,Akio Kanazawa,Akihiro Sekine,Susumu Saito,Tatsuhiko Tsunoda,Daisuke Koya,Tetsuya Babazono,Yasushi Tanaka,Masafumi Matsuda,Koichi Kawai,Tomohiro Iiizumi,Masahito Imanishi,Toshihiro Shinosaki,Toru Yanagimoto,Minoru Ikeda,Shigeki Omachi,Atsunori Kashiwagi,Kohei Kaku,Yasuhiko Iwamoto,R. Kawamori,Ryuichi Kikkawa,Masatoshi Nakajima,Yusuke Nakamura,Shiro Maeda +24 more
TL;DR: Results indicate that ELMO1 is a novel candidate gene that both confers susceptibility to DN and plays an important role in the development and progression of this disease.

Association of the Gene Encoding Wingless-Type Mammary Tumor Virus Integration-Site Family Member 5B (WNT5B) with Type 2 Diabetes
Akio Kanazawa,Syuuichi Tsukada,Akihiro Sekine,Tatsuhiko Tsunoda,Atsushi Takahashi,Atsunori Kashiwagi,Yasushi Tanaka,Tetsuya Babazono,Masafumi Matsuda,Kohei Kaku,Yasuhiko Iwamoto,Ryuzo Kawamori,Ryuichi Kikkawa,Yusuke Nakamura,Shiro Maeda +14 more
TL;DR: The results indicate that the WNT5B gene may contribute to conferring susceptibility to type 2 diabetes and may be involved in the pathogenesis of this disease through the regulation of adipocyte function.

Wnt5b partially inhibits canonical Wnt/β-catenin signaling pathway and promotes adipogenesis in 3T3-L1 preadipocytes
Akio Kanazawa,Shuichi Tsukada,Masumi Kamiyama,Toru Yanagimoto,Masatoshi Nakajima,Shiro Maeda +5 more
TL;DR: The overexpression of Wnt5b was able to inhibit Wnt3a-induced activation of the canonical Wnt/beta-catenin pathway as evidenced by the reduced translocation of beta-Catenin into the nucleus.
