When tissue cells are cultured on very thin sheets of cross-linked silicone fluid, the traction forces the cells exert are made visible as elastic distortion and wrinkling of this substratum. Around explants this pattern of wrinkling closely resembles the "center effects" long observed in plasma clots and traditionally attributed to dehydration shrinkage.

Silicone rubber substrata: a new wrinkle in the study of cell locomotion

The locomotion of fibroblasts in culture. IV. Electron microscopy of the leading lamella.

A technique for exciting fluorescence exclusively from regions of contact between cultured cells and the substrate is presented. The technique utilizes the evanescent wave of a totally internally reflecting laser beam to excite only those fluorescent molecules within one light wavelength or less of the substrate surface. Demonstrations of this technique are given for two types of cell cultures: rat primary myotubes with acetylcholine receptors labeled by fluorescent alpha-bungarotoxin and human skin fibroblasts labeled by a fluorescent lipid probe. Total internal reflection fluorescence examination of cells appears to have promising applications, including visualization of the membrane and underlying cytoplasmic structures at cell-substrate contacts, dramatic reduction of autofluorescence from debris and thick cells, mapping of membranes topography, and visualization of reversible bound fluorescent ligands at membrane receptors.

Cell-substrate contacts illuminated by total internal reflection fluorescence.

The synthetic peptide Gly-Arg-Gly-Asp-Tyr (GRGDY), which contains the RGD sequence of several adhesion molecules, was covalently grafted to the surface of otherwise poorly adhesive glass substrates and was used to determine the minimal number of ligand-receptor interactions required for complete spreading of human foreskin fibroblasts. Well-defined adhesion substrates were prepared with GRGDY between 10(-3) fmol/cm2 and 10(4) fmol/cm2. As the adhesion ligand surface concentration was varied, several distinct morphologies of adherent cells were observed and categorized. The population of fully spread cells at 4 h reached a maximum at 1 fmol/cm2, with no further increases up to 10(4) fmol/cm2. Although maximal cell spreading was obtained at 1 fmol/cm2, focal contacts and stress fibers failed to form at RGD surface concentrations below 10 fmol/cm2. The minimal peptide spacings obtained in this work correspond to 440 nm for spreading and 140 nm for focal contact formation, and are much larger than those reported in previous studies with adsorbed adhesion proteins, adsorbed RGD-albumin conjugates, or peptide-grafted polyacrylamide gels. Vitronectin receptor antiserum specific for integrin alpha V beta 3 blocked cell adhesion and spreading on substrates containing 100 fmol/cm2 of surface-bound GRGDY, while fibronectin receptor antiserum specific for alpha 5 beta 1 did not. Furthermore, alpha V beta 3 was observed to cluster into focal contacts in spread cells, but alpha 5 beta 1 did not. It was thus concluded that a peptide-to-peptide spacing of 440 nm was required for alpha V beta 3-mediated cellular spreading, while 140 nm was required for alpha V beta 3-mediated focal contact formation and normal stress fiber organization in human foreskin fibroblasts; these spacings represent much fewer ligands than were previously thought to be required.

Integrin-based adhesion has served as a model for studying the central role of adhesion in migration. In this article, we outline modes of migration, both integrin-dependent and -independent in vitro and in vivo. We next discuss the roles of adhesion contacts as signaling centers and linkages between the ECM and actin that allows adhesions to serve as traction sites. This includes signaling complexes that regulate migration and the interplay among adhesion, signaling, and pliability of the substratum. Finally, we address mechanisms of adhesion assembly and disassembly and the role of adhesion in cellular polarity.

Integrins in Cell Migration

An optical technique for measuring the thickness of thin films has been adapted and evaluated for studying the structure of the adhesion of cells to glass in tissue culture. This technique, which is termed interference reflection microscopy, has been used to study embryonic chick heart fibroblasts. These findings have been observed: in normal culture medium the closest approach of the cell surface to substrate in its adhesions is ca . 100 A, much of the cell surface lying farther away; chemical treatments which bring the cell surface to near its charge reversal point reduce the closest approach of adhesions to ca . 100 A; high osmotic concentration of a non-polar substance, i.e . sucrose, does not affect the distance between cell and substrate in the adhesions. In addition, optical evidence indicates that there is no extracellular material between cell and glass in the adhesions. When cells de-adhere from glass, they appear not to leave fragments behind. The adhesive sites in these fibroblasts appear to be confined to the edge of the side of the cell facing the substrate and to the pseudopods. The significance of this is discussed in relation to the phenomenon of contact inhibition. Evidence is presented that the mechanism of cell adhesion does not involve calcium atoms binding cells to substrate by combining with carboxyl groups on cell surface, substrate, and with a cement substance. Osmium tetroxide fixation results in a final separation of 100 to 200 A between cell and substrate: there are reasons for thinking that this fairly close approach to the condition in life is produced as an artefact. The results can be accounted for only in terms of the action of electrostatic repulsive forces and an attractive force, probably the van der Waals—London forces. Biological arguments suggest that these results are equally applicable for cell-to-cell adhesions.
