Abstract: Dendritic cells (DCs) are professional antigen-presenting cells that provide a critical link between the innate and adaptive immune responses. The genetic program required for differentiation of DCs from their hematopoietic precursors is controlled by both cytokines and transcription factors. The signals transduced from cytokines recruit specific transcription factors, enabling the expression of a distinct transcriptome that is required for specification of different DC lineages. The establishment of a distinct transcriptome also depends on chromatin modifications associated with critical cis elements of lineage-specific genes. In this review, recent advances in the understanding of the transcriptional network governing DC lineage specification are summarized, along with current views of the dynamic DC epigenome.

Efficient presentation of soluble antigen by cultured human dendritic cells is maintained by granulocyte/macrophage colony-stimulating factor plus interleukin 4 and downregulated by tumor necrosis factor alpha.
TL;DR: Cultured DCs are as efficient as antigen-specific B cells in presenting tetanus toxoid (TT) to specific T cell clones and their efficiency of antigen presentation can be further enhanced by specific antibodies via FcR- mediated antigen uptake.

TL;DR: The methodology for inducing dendritic cell growth that was recently described for mouse blood now has been modified to MHC class II- negative precursors in marrow, and this feature should prove useful for future molecular and clinical studies of this otherwise trace cell type.
Development of Monocytes, Macrophages, and Dendritic Cells
TL;DR: The current understanding of myeloid lineage development is reviewed and the developmental pathways and cues that drive differentiation are described, which are central to the development of immunologic memory and tolerance in mice.
Mouse and human dendritic cell subtypes

TL;DR: The dynamics of the DC network in response to microbial invasion is studied, because many DC subtypes arise from separate developmental pathways, and their development and function are modulated by exogenous factors.
