The synthetic retinoid (4-hydroxyphenyl)retinamide decreases collagen expression in vitro and in the tight-skin mouse
23
TL;DR: It is shown that (4-hydroxyphenyl)retinamide decreases alpha 1(I) collagen messenger RNA and transcription in cultured cells, and decreases collagen levels in the dermis of tight-skin mice.

Abstract: Objective. We documented the effect of the retinoid (4-hydroxyphenyl)retinamide on collagen expression in a tissue culture and in an animal model of scleroderma.
Methods. We used RNA analysis, chloramphenicol acetyltransferase assays, organ culture, and histologic evaluation.
Results. We showed that (4-hydroxyphenyl)retinamide decreases αl(I) collagen messenger RNA and transcription in cultured cells, and decreases collagen levels in the dermis of tight-skin mice.
Conclusion. These results provide a basis for further experiments to address the efficacy of (4-hydroxyphenyl)retinamide in the treatment of scleroderma.

Halofuginone: A Novel Antifibrotic Therapy
Mark Pines,Arnon Nagler +1 more
TL;DR: The ability of extremely low concentrations of halofuginone to inhibit collagen alpha 1(I) synthesis specifically and transiently at the transcriptional level suggests that this material fulfills the criteria for a successful and effective anti-fibrotic therapy.

Inhibition of collagen synthesis and changes in skin morphology in murine graft-versus-host disease and tight skin mice: effect of halofuginone
TL;DR: The low concentration of halofuginone needed to prevent collagen deposition in fibrotic skin without affecting body weight suggests that halofUGinone may serve as a novel and promising anti-fibrotic therapy.

Pathophysiology of systemic sclerosis: state of the art in 2014.
TL;DR: A number of new experimental models have been set up, that should help to understand the disease pathogenesis and test new therapeutic targets and Plasmacytoid dendritic cells seem to play a major role in the pathogenesis of SSc through the secretion of CXCL4.

The pathogenesis of cutaneous fibrosis.
Bin Liu,M. K. Connolly +1 more
TL;DR: This work examines various molecular events in scleroderma that may lead to fibroblast activation, and proposes a new model to explain the persistence of such activation by sclerodma fibroblasts in the apparent absence of exogenous stimuli.

N-(4-hydroxyphenyl)retinamide (Fenretinide) in combination with retinoic acid enhances differentiation and retinoylation of proteins.
TL;DR: 4-HPR may be a member of a class of retinoids that are active because they displace RA from extracellular and intracellular sites or because they inhibit RA catabolism, and may have utility in the clinic in combination with RA.

A technique for radiolabeling DNA restriction endonuclease fragments to high specific activity
TL;DR: A technique for conveniently radiolabeling DNA restriction endonuclease fragments to high specific activity is described, and these "oligolabeled" DNA fragments serve as efficient probes in filter hybridization experiments.
