Abstract: Two Photon Excited Fluorescence (TPEF) allows for high-resolution non-invasive retinal imaging, and benefits from guidance of Optical Coherence Tomography (OCT) and Sensorless Adaptive Optics (SAO). We present our progress on the use of SAO with OCT and TPEF for imaging the retina in mice.

Two-Photon Laser Scanning Fluorescence Microscopy
TL;DR: The fluorescence emission increased quadratically with the excitation intensity so that fluorescence and photo-bleaching were confined to the vicinity of the focal plane as expected for cooperative two-photon excitation.

TL;DR: To increase fluorescence intensity and reduce the requisite laser power, this work modulated ultrashort laser pulses with high-order dispersion compensation and applied sensorless adaptive optics and custom image recovery software and observed an over 300% increase in fluorescence of endogenous retinal fluorophores when laser pulses were shortened from 75 fs to 20 fs.
