Multiplex semi-quantitative reverse transcriptase-polymerase chain reaction of low abundance neuronal mRNAs.
72
TL;DR: A RT-PCR assay which allows the simultaneous detection and semi-quantitation of several transcripts (multiplex) and normalization with respect to HPRT cDNA allows to estimate the relative abundance of each target mRNA.

Increased BMP6 levels in the brains of Alzheimer’s disease patients and APP transgenic mice are accompanied by impaired neurogenesis
Leslie Crews,Anthony Adame,Christina Patrick,Alexandra DeLaney,Emiley Pham,Edward Rockenstein,Lawrence A. Hansen,Eliezer Masliah +7 more
TL;DR: It is suggested that Aβ-associated increases in BMP6 expression in AD may have deleterious effects on neurogenesis in the hippocampus, and therapeutic approaches could focus on normalization of B MP6 levels to protect against AD-related neurogenic deficits.

Proteasome inhibition enhances the induction and impairs the maintenance of late-phase long-term potentiation.
TL;DR: It is shown here that inhibition of the proteasome enhances the induction of L-LTP, but inhibits its maintenance, and that proteasomesome-mediated proteolysis has different roles during the induction and the maintenance of L -LTP.

Altered midbrain dopaminergic neurotransmission during development in an animal model of ADHD
Damiana Leo,Eleonora Sorrentino,Floriana Volpicelli,Maria Eyman,Dario Greco,Davide Viggiano,Umberto di Porzio,Carla Perrone-Capano +7 more
TL;DR: It is suggested that down-regulation of DA neurotransmission occurs in the midbrain of SHR in a developmentally regulated temporal window during postnatal development, thus strengthening the hypodopaminergic hypothesis in the pathogenesis of ADHD.

Regulation of Editing and Expression of Glutamate α-Amino-Propionic-Acid (AMPA)/Kainate Receptors by Antidepressant Drugs
Alessandro Barbon,Maurizio Popoli,Luca La Via,Stefania Moraschi,Ivan Vallini,Daniela Tardito,Ettore Tiraboschi,Laura Musazzi,R. Giambelli,Massimo Gennarelli,Giorgio Racagni,Sergio Barlati +11 more
TL;DR: Results point to specific and regionally discrete changes in the expression and editing level of glutamate receptors and, in particular, to a selective reduction of conductance for GluR3-containing receptors following treatment with antidepressant drugs.

Bdnf gene is a downstream target of Nurr1 transcription factor in rat midbrain neurons in vitro.
Floriana Volpicelli,Massimiliano Caiazzo,Dario Greco,Claudia Consales,Luigi Leone,Carla Perrone-Capano,Luca Colucci D’Amato,Umberto di Porzio +7 more
TL;DR: It is demonstrated that following depolarization the hyperexpression of Nurr1 and the brain derived neurotrophic factor (BDNF) are phospholipase C and protein kinase C‐dependent and it is suggested that Nurr 1 might also influence the development and the function of midbrain dopaminergic neurons via direct regulation of Bdnf expression.

Single-step method of RNA isolation by acid guanidinium thiocyanate-phenol-chloroform extraction
TL;DR: A new method of total RNA isolation by a single extraction with an acid guanidinium thiocyanate-phenol-chloroform mixture is described, providing a pure preparation of undegraded RNA in high yield and can be completed within 4 h.

Prevention of pre-PCR mis-priming and primer dimerization improves low-copy-number amplifications.
TL;DR: Improved amplification performance is evident for target copy numbers below approximately 10(3), and when mis-priming is negligible, the procedural improvement still suppresses putative primer oligomerization.

General concepts for PCR primer design.
TL;DR: The purpose of this chapter is to explain basic rules of oligonucleotide primer design, and how the information deduced by primer design software can be rationally interpreted and manipulated to fit experimental needs.
