Abstract: PURPOSE:To more enhance the frequency of transforming a yeast according to simple treatment by culturing a yeast cell according to a conventional method, then subjecting the cultured yeast to suspending treatment with alkali metallic ions, subsequently heat-treating the yeast under specific conditions, bringing the objective DNA into contact with the yeast and transducing the DNA thereinto CONSTITUTION:In a method for transforming a yeast by treating the yeast cell with alkali metallic ions, bringing the objective DNA into contact with the treated yeast cell and transducing the DNA into the yeast cell, a yeast (Saccharomyces cerevisiae MC16 strain) is inoculated into a culture medium and precultured at 30 degC When a stationary phase is attained, the precultured solution is inoculated into a new culture medium and resubjected to shaking culture at 30 degC The fungal cell is aseptically collected, subjected to suspending treatment with alkali metallic ions such as lithium acetate, then mixed with the objective DNA such as a plasmid and heat-treated at 43-47 degC for 1-15min The DNA is transduced into the yeast to enhance the frequency of transformation and transform the yeast according to simple treatment
