TL;DR: It is shown that immune competence for even stringent antigens such as HY can be restored in the absence of thymic function and IL-7 is identified as a potent modulator ofThymic-independent T-cell regeneration.

The Many Faces of IL-7: From Lymphopoiesis to Peripheral T Cell Maintenance
Terry J. Fry,Crystal L. Mackall +1 more
TL;DR: IL-7 plays critical and nonredundant roles in both T cell lymphopoiesis and in maintaining and restoring peripheral T cell homeostasis.

Harnessing the biology of IL-7 for therapeutic application
TL;DR: The biology of IL-7 and the results of its clinical use that are available so far are summarized to provide a perspective on the opportunities for clinical application of this cytokine.
Interleukin-7: from bench to clinic.
Terry J. Fry,Crystal L. Mackall +1 more
TL;DR: The complete set of physiologic roles for this cytokine, especially those involving lymphocyte homeostasis, have only recently been elucidated, after the initial descriptions of effects on B-cell precursors.

Thymic involution and immune reconstitution
Heather E. Lynch,Gabrielle L. Goldberg,Ann P. Chidgey,Marcel R.M. van den Brink,Richard L. Boyd,Gregory D. Sempowski +5 more
TL;DR: Progress is presented on the exploitation of thymosuppressive and thymostimulatory pathways using factors such as keratinocyte growth factor, interleukin 7 or sex steroid ablation for therapeutic thymus restoration and peripheral immune reconstitution in adults.

T cell immune reconstitution following lymphodepletion.
TL;DR: Insight has permitted the manipulation of the immune system to maximize anti-tumor immunity through lymphopenia and led to an appreciation of mechanisms that underlie graft versus host disease.

IL-7 increases both thymic-dependent and thymic-independent T-cell regeneration after bone marrow transplantation
TL;DR: The administration of exogenous IL-7, and to a lesser extent IL-2, abrogates the down-regulation in antigen-driven peripheral expansion that occurs in thymus-bearing hosts after BMT, suggesting that one mechanism by which T-cell-depleted hosts may support antigen- driven T-cells expansion in vivo is via an increased availability of T- cell-active cytokines to support clonal expansion.
