Interleukin-18 regulates both Th1 and Th2 responses.
1.3K
TL;DR: IL-18 is a potent proinflammatory cytokine that has pathophysiological roles in several inflammatory conditions, and in principle, IL-18 enhances the IL-12-driven Th1 immune responses, but it can also stimulate Th2 immune responses in the absence of IL- 12.

Abstract: Although interleukin-18 is structurally homologous to IL-1 and its receptor belongs to the IL-1R/Toll-like receptor (TLR) superfamily, its function is quite different from that of IL-1. IL-18 is produced not only by types of immune cells but also by non-immune cells. In collaboration with IL-12, IL-18 stimulates Th1-mediated immune responses, which play a critical role in the host defense against infection with intracellular microbes through the induction of IFN-gamma. However, the overproduction of IL-12 and IL-18 induces severe inflammatory disorders, suggesting that IL-18 is a potent proinflammatory cytokine that has pathophysiological roles in several inflammatory conditions. IL-18 mRNA is expressed in a wide range of cells including Kupffer cells, macrophages, T cells, B cells, dendritic cells, osteoblasts, keratinocytes, astrocytes, and microglia. Thus, the pathophysiological role of IL-18 has been extensively tested in the organs that contain these cells. Somewhat surprisingly, IL-18 alone can stimulate Th2 cytokine production as well as allergic inflammation. Therefore, the functions of IL-18 in vivo are very heterogeneous and complicated. In principle, IL-18 enhances the IL-12-driven Th1 immune responses, but it can also stimulate Th2 immune responses in the absence of IL-12.

Human Vδ2 T Cells and Their Versatility for Immunotherapeutic Approaches
TL;DR: In this paper , major characteristics of the most abundant circulating γδ subpopulation, Vδ2 T cells, their immunotherapeutic potential, recent advances in expansion protocols, their preclinical and clinical applications for several infectious diseases and malignancies, and how additional modulation could enhance their therapeutic potential.
CAR T Cells Secreting IL18 Augment Antitumor Immunity and Increase T Cell Proliferation and Costimulation
Biliang Hu,Jiangtao Ren,Yanping Luo,Brian Keith,Regina M. Young,John Scholler,Yangbing Zhao,Carl H. June +7 more
TL;DR: This study demonstrates that ectopic expression of IL18 in CART cells caused significant T cell proliferation in vitro and in vivo, and enhanced antitumor effects in xenograft models, and supports human IL18 as a T cell costimulatory cytokine for fueling CART therapy.

Lan Zhi Wang,Ya Ping Xu,Yi Lian Zhou,Ze Peng Liu,Bo Li,Wen Bin Gu,Xiao Feng Zhao,Wei Ren Dong,Miao An Shu +8 more
TL;DR: The expression profiles of AdIL-18s were investigated under the challenges of Aeromonas hydrophila, Staphylococcus ureae and Poly (I:C) respectively, and the results suggested that AdILs were involved in the immune responses against both bacterial and viral infections.

Reconstitution of the immunological defence and Candida albicans infection in oral mucosa of HIV+ patients under HAART
Marinho Del Santo
- 01 Jan 2016
TL;DR: The HIV infection is a worldwide spread disease which with the HAART (highly active antiretroviral therapy) application has became a chronicle disease and the sensitive control to balance immunological innate and immunological acquired actions is complex.

Elevated systemic interleukin-18 in multiple injured patients is not related to clinical outcome.
TL;DR: The data demonstrate elevated plasma levels of IL-18 after severe trauma, which were significantly increased in polytraumatized patients compared to healthy donors and survivors and presented significantly higher than expected in survivors.
