Insulin in the Brain: Sources, Localization and Functions
287
TL;DR: This review summarizes the current status of knowledge of the peripheral and central sources of insulin in the brain, site-specific expression of IRs, and also neurophysiological functions of insulin including the regulation of food intake, weight control, reproduction, and cognition and memory formation and considers the neuromodulatory and neurotrophic effects of insulin.

Abstract: Historically, insulin is best known for its role in peripheral glucose homeostasis, and insulin signaling in the brain has received less attention. Insulin-independent brain glucose uptake has been the main reason for considering the brain as an insulin-insensitive organ. However, recent findings showing a high concentration of insulin in brain extracts, and expression of insulin receptors (IRs) in central nervous system tissues have gathered considerable attention over the sources, localization, and functions of insulin in the brain. This review summarizes the current status of knowledge of the peripheral and central sources of insulin in the brain, site-specific expression of IRs, and also neurophysiological functions of insulin including the regulation of food intake, weight control, reproduction, and cognition and memory formation. This review also considers the neuromodulatory and neurotrophic effects of insulin, resulting in proliferation, differentiation, and neurite outgrowth, introducing insulin as an attractive tool for neuroprotection against apoptosis, oxidative stress, beta amyloid toxicity, and brain ischemia.

The Impact of Vitamin D Supplementation on Neurodegeneration, TNF-α Concentration in Hypothalamus, and CSF-to-Plasma Ratio of Insulin in High-Fat-Diet-Induced Obese Rats.
Ghazaleh Nameni,Ghazaleh Hajiluian,Parviz Shahabi,Mahdieh Abbasalizad Farhangi,Mehran Mesgari-Abbasi,Mohammad-Reza Hemmati,Seyed Mahdi Vatandoust +6 more
TL;DR: Vitamin D improved brain insulin homeostasis and modulated food intake and body weight in high-fat-diet-induced obese rats and had no significant effect on degenerated neurons and TNF-α concentration in the hypothalamus.

Effects of exercise on brain functions in diabetic animal models.
TL;DR: This review describes how exercise corrects alterations in the brain by DM such as increased adiposity, inflammation, hormonal dysregulation, uncontrolled hyperphagia, insulin and leptin resistance, and dysregulation of neurotransmitters and declined neurogenesis in the hippocampus in animal models.

- 01 Jan 2014
TL;DR: This book will elaborate on the mitochondrial mechanisms regulated by the ghrelin receptor, its role in feeding and drug addictive mechanisms, memory, sleep and arousal, and the potential of this receptor as a target for the treatment of neurological disorders including Parkinson’s disease, epilepsy, anxiety and depression.

Intranasal Insulin Prevents Anesthesia-induced Cognitive Impairments in Aged Mice.
Xing Li,Xiaoqin Run,Zhen Wei,Kuan Zeng,Zhihou Liang,Fang Huang,Dan Ke,Qun Wang,Jian-Zhi Wang,Rong Liu,Bin Zhang,Xiaochuan Wang +11 more
TL;DR: Intranasal insulin administration might be a promising therapy to prevent anesthesiainduced cognitive deficit in elderly individuals.

Insulin resistance and cognitive test performance in elderly adults: National health and nutrition examination survey (NHANES).
TL;DR: Insulin resistance as measured by HOMA-IR was independently associated with lower cognitive test performance score among elderly participants aged ≥60years and longitudinal studies are needed to test the mechanism and the causal relationship.

TL;DR: Mice created with a neuron-specific disruption of the IR gene showed increased food intake, and both male and female mice developed diet-sensitive obesity with increases in body fat and plasma leptin levels, mild insulin resistance, elevated plasma insulin levels, and hypertriglyceridemia.

Insulin-mediated skeletal muscle vasodilation is nitric oxide dependent. A novel action of insulin to increase nitric oxide release.
TL;DR: Insulin vasodilation of skeletal muscle vasculature most likely occurs via increasing EDNO synthesis/release, and insulin appears to be a novel modulator of the EDNO system.
