Abstract: We previously reported that the inhibition of catalase and glutathione peroxidase activities by treatment with 3-amino-1,2,4-triazole (ATZ) and mercaptosuccinic acid evoked sustained increases in the levels of reactive oxygen species and apoptosis in rat primary hepatocytes. Apoptosis was accompanied by increased expression of BimEL, following activation of extracellular signal-regulated kinase. The aim of this study was to characterize the mechanism underlying hepatocyte apoptosis by identifying the transcription factor that induces BimEL expression. The bim promoter region was cloned into a promoterless-luc vector, and promoter activity was monitored by a luciferase assay. The luciferase activity increased in the presence of ATZ + mercaptosuccinic acid. Pretreatment with a MEK inhibitor, U0126, or an antioxidant, vitamin C, suppressed the promoter activity. Furthermore, ATZ + mercaptosuccinic acid-induced luciferase activity was attenuated by mutation of the activator protein-1 binding site in the bim promoter region. The amounts of total and phosphorylated c-Fos increased over time in the presence of ATZ + mercaptosuccinic acid, whereas the amounts of total and phosphorylated c-Jun remained unchanged. Chromatin immunoprecipitation revealed that both c-Fos and c-Jun localized to the activator protein-1-binding site in the bim promoter region. BimEL expression and hepatocyte apoptosis were suppressed by knockdown of c-Fos and c-Jun, respectively. These results indicate that increases in c-Fos following extracellular signal-regulated kinase activation are critical for BimEL upregulation and apoptosis.

Hydrogen sulfide exposure triggers chicken trachea inflammatory injury through oxidative stress-mediated FOS/IL8 signaling.
TL;DR: H2S induced oxidative stress leads to tracheal inflammation through FOS/IL8 signaling, leading to excessive mucus secretion and absence of cilia, and new insights are provided for unveiling the biological effects of H2S in vivo and in vitro.

Tributyltin induces oxidative stress and neuronal injury by inhibiting glutathione S-transferase in rat organotypic hippocampal slice cultures
TL;DR: This study is the first report to show the link between neuronal oxidative injury and the GST inhibition elicited by TBT, indicating that GST inhibition could contribute, at least in part, to the neuronal cell death induced by T BT in hippocampal slices.

c-Fos is a mechanosensor that regulates inflammatory responses and lung barrier dysfunction during ventilator-induced acute lung injury
TL;DR: Wang et al. as discussed by the authors investigated whether T-5224 attenuates VILI in rats by inhibiting inflammation and apoptosis, which is associated with the anti-inflammatory and anti-apoptotic effects of T5224.
Transcriptional Regulation of Renal Dopamine D1 Receptor Function During Oxidative Stress
TL;DR: Tempol mitigates oxidative stress, blocks AP1–SP3 activation, and prevents D1R dysfunction and hypertension in both human kidney cells and rats and shows that oxidative stress via AP1- or SP3-specific siRNA suppresses D 1R transcription and function.

Purified transcription factor AP-1 interacts with TPA-inducible enhancer elements
TL;DR: It is demonstrated that multiple synthetic copies of the consensus AP-1-binding site can act as TPA-inducible enhancers in various plasmid constructs after transfection into HeLa cells, suggesting that AP- 1 is a transcription factor that functions by interacting with a specific enhancer element, and that its activities may be modulated by treatment of cells with TPA.
