Abstract: by Rachel Harding Unlike most other vertebrates, zebrafish are able to regenerate many tissues including retina. Light-induced photoreceptor cell death causes inner nuclear layer Müller glia to dedifferentiate, reenter the cell cycle and produce proliferating progenitor cells. These progenitor cells continue to proliferate and migrate to the appropriate retinal layer and differentiate into functional photoreceptors. While a number of genes essential to retinal regeneration have been identified, the mechanisms regulating these genes have been little studied. Many post-transcriptional regulatory mechanisms exist in the cell including miRNAs, short RNA sequences encoded in the genome. To determine if miRNAs play a role in retinal regeneration, I reduced global miRNA expression by Dicer knockdown, an enzyme essential to miRNA maturation. Dicer knockdown prior to light damage lead to decreased proliferation in regenerating retinas without affecting the number of Müller glia during retinal regeneration or cell death. These studies demonstrate an essential role

TL;DR: Green fluorescent protein and BrdU-mediated lineage tracing combined with retinal gene expression analysis indicated that α1T-expressing Müller glia were capable of generating retinal neurons and glia, and strongly suggest α1 T-expressed Mủller gla dedifferentiate and mediate regeneration of the injured zebrafish retina.

TL;DR: Although development is initially more advanced within the ventronasal region between 50 and 60 hpf, development across the retina rapidly equilibrates such that it is relatively comparable within all quadrants of the central retina by 70 hpf.

MicroRNA expression profiling of single whole embryonic stem cells
TL;DR: It is demonstrated that it is possible to analyse miRNA in single cells by using a real-time PCR-based 220-plex miRNA expression profiling method, which is critical for elucidating the diverse roles of miRNAs.
GFAP transgenic zebrafish.
TL;DR: GFP expression in glia co-localized with anti-GFAP antibodies, but did not co- localize with the neuronal antibodies HuC/D or calretinin in mature neurons, with moderate expression in the enteric nervous system and weaker levels in the olfactory sensory placode and otic capsule.
