The Role of Astrocytic Calcineurin Activation and Downstream Signaling in Neurodegenerative Diseases
Melanie M. Pleiss
- 01 Jan 2016
TL;DR: A custom antibody is created that selectively identifies a proteolyzed, constitutively active CN fragment and blockade of CN/Cx43 interactions reduced hemichannel permeability in primary astrocytes following treatment with IL-1β, suggesting that aberrant CN activation in astroCytes may negatively affect neurons via interactions with Cx43-containing hemichannels.
Role of Actin-mediated Motility of Peripheral Astrocytic Processes in Synaptic Function
Dmitry Molotkov
- 12 Jun 2014
TL;DR: In vivo methods to probe astrocytic morphological and functional changes and methodological approaches to study neuro-glial interactions .........................................................................
Astroglial connexins and cognition: memory formation or deterioration?
TL;DR: The memory formation as well as memory deteriorating functions of astrocytes-located connexins in memory disorders of different etiology with possible mechanisms are described.
Cannabinoids prevent the amyloid β-induced activation of astroglial hemichannels: A neuroprotective mechanism
Rosario Gajardo-Gómez,Valeria C. Labra,Carola J. Maturana,Kenji F. Shoji,Cristian A. Santibañez,Juan C. Sáez,Christian Giaume,Juan A. Orellana +7 more
TL;DR: It is demonstrated that synthetic and endogenous cannabinoids (CBs) reduce the opening of astrocyte Cx43 hemichannels evoked by activated microglia or inflammatory mediators and fully abolished the Aβ‐induced release of excitotoxic glutamate and ATP associated to astroglial hemichannel activity, as well as neuronal damage in hippocampal slices exposed to Aβ.
The Role of Connexin Hemichannels in Inflammatory Diseases
TL;DR: It is increasingly clear that CxHC can be induced to open by pathogen-associated molecular patterns, and this review provides an update on the progress in the understanding of Cx HC, with a focus on the role of these channels in inflammatory diseases.

Alzheimer's Disease: Genes, Proteins, and Therapy
TL;DR: Evidence that the presenilin proteins, mutations in which cause the most aggressive form of inherited AD, lead to altered intramembranous cleavage of the beta-amyloid precursor protein by the protease called gamma-secretase has spurred progress toward novel therapeutics and provided discrete biochemical targets for drug screening and development.

TL;DR: By better understanding AD inflammatory and immunoregulatory processes, it should be possible to develop anti-inflammatory approaches that may not cure AD but will likely help slow the progression or delay the onset of this devastating disorder.

ATP mediates rapid microglial response to local brain injury in vivo
Dimitrios Davalos,Jaime Grutzendler,Jaime Grutzendler,Guang Yang,Jiyun Kim,Yi Zuo,Steffen Jung,Dan R. Littman,Michael L. Dustin,Wen-Biao Gan +9 more
TL;DR: Extracellular ATP regulates microglial branch dynamics in the intact brain, and its release from the damaged tissue and surrounding astrocytes mediates a rapid microglia response towards injury.
Molecular dissection of reactive astrogliosis and glial scar formation.
TL;DR: Developments in the signaling mechanisms that regulate specific aspects of reactive astrogliosis are reviewed and the potential to identify novel therapeutic molecular targets for diverse neurological disorders is highlighted.

Glial and neuronal control of brain blood flow.
David Attwell,Alastair M. Buchan,Serge Charpak,Martin Lauritzen,Brian A. MacVicar,Eric A. Newman +5 more
TL;DR: It is now recognized that neurotransmitter-mediated signalling has a key role in regulating cerebral blood flow, that much of this control is mediated by astrocytes, that oxygen modulates blood flow regulation, and that blood flow may be controlled by capillaries as well as by arterioles.
