Abstract: Cancer stem cells (CSCs) play a critical role in the cancer metastasis and account for tumor heterogeneity. Growing evidence indicates that the CSC phenotypes are related to the tumor microenvironment. In this study, we report that the gradient of mechanical stresses guides the spatial patterning of the expression of CD44 and Yes-associated protein (YAP) in the geometrically confined multicellular sheets. Our study shows that the cytoskeletal contraction regulates the expression of CD44 through the translocation of YAP into the nucleus. The results demonstrate that geometric confinement and mechanical stresses are the regulators in the spatial patterning of CSC. It may help to understand the relationship between the tumor microenvironment and oncogenesis.

Spontaneous formation and spatial self-organization of mechanically induced mesenchymal-like cells within geometrically confined cancer cell monolayers
01 Feb 2022
TL;DR: In this article , the authors developed a micropattern-based cell monolayer model to investigate the regulation of mechanics heterogeneity on the cell phenotypic switch and reveal that mechanics heterogeneity itself is enough to spontaneously induce the emergence of mesenchymal-like phenotype and asymmetrical activation of TGF-β-SMAD signaling.

Mechanical Constraints in Tumor Guide Emergent Spatial Patterns of Glioblastoma Cancer Stem Cells
01 Oct 2023
TL;DR: Mechanical constraints in glioblastoma microenvironment regulate tumor heterogeneity and cancer stem cell emergence, with CSCs exhibiting stem-like properties and spatially patterning in peripheral regions due to mechanical stress gradients and Piezo1-RhoA-focal adhesion signaling.

Pressure loading regulates the stemness of liver cancer stem cells via YAP/BMF signaling axis
Di Ma,Rui Liang,Qing Luo,Guanbin Song +3 more
TL;DR: Pressure loading regulates liver cancer stem cell stemness via YAP/BMF signaling axis, with increased pressure upregulating CSC markers and tumorigenic potential, but excessive pressure suppressing these characteristics, highlighting a potential therapeutic target for CSCs.
Matrix stiffening facilitates stemness of liver cancer stem cells by YAP activation and BMF inhibition
Di Ma,Qing Luo,Guanbin Song +2 more
TL;DR: The regulatory mechanism of heterogeneous matrix stiffness on CSC stemness is unraveled and a novel therapeutic strategy for eradicating CSCs and improving the efficiency of HCC treatment is provided.
Geometric constraint-triggered collagen expression mediates bacterial-host adhesion

TL;DR: It is revealed that interactions between bacteria and spatially constrained cell monolayer exhibit strong spatial heterogeneity, and that bacteria tend to adhere to these cells near the outer edges of confined monolayers.

Matrix elasticity directs stem cell lineage specification.
TL;DR: Naive mesenchymal stem cells are shown here to specify lineage and commit to phenotypes with extreme sensitivity to tissue-level elasticity, consistent with the elasticity-insensitive commitment of differentiated cell types.

Stem cells, cancer, and cancer stem cells
TL;DR: Stem cell biology has come of age: Unequivocal proof that stem cells exist in the haematopoietic system has given way to the prospective isolation of several tissue-specific stem and progenitor cells, the initial delineation of their properties and expressed genetic programmes, and the beginnings of their utility in regenerative medicine.
Human acute myeloid leukemia is organized as a hierarchy that originates from a primitive hematopoietic cell

TL;DR: It is demonstrated that the cell capable of initiating human AML in non-obese diabetic mice with severe combined immunodeficiency disease (NOD/SCID mice) — termed the SCID leukemia-initiating cell, or SL-IC — possesses the differentiate and proliferative capacities and the potential for self-renewal expected of a leukemic stem cell.

The extracellular matrix at a glance
TL;DR: The extracellular matrix is the non-cellular component present within all tissues and organs, and provides not only essential physical scaffolding for the cellular constituents but also initiates crucial biochemical and biomechanical cues that are required for tissue development.

TL;DR: A workshop was convened by the AACR to discuss the rapidly emerging cancer stem cell model for tumor development and progression, and participants were charged with evaluating data suggesting that cancers develop from a small subset of cells with self-renewal properties analogous to organ regeneration.
