TL;DR: CD4(+) T cells from patients deficient in IL-12Rβ1, TYK2, STAT1, and STAT3 are examined to further explore the pathways involved in human Tfh cell differentiation and suggest that defective T fh cell development and/or function contributes to the humoral defects observed in STAT3-deficient patients.

Transcription Factor STAT3 Serves as a Negative Regulator Controlling IgE Class Switching in Mice.
Paul Dascani,Chuanlin Ding,Xiangyu Kong,Xiangyu Kong,David Tieri,Xiaoling Hu,Huang-Ge Zhang,Daisuke Kitamura,Roberto Bolli,Eric C. Rouchka,Jun Yan +10 more
- 01 Dec 2018
TL;DR: A negative role for STAT3 in regulating class switching of the GC B cells from the IgG1 to the IgE producing state is suggested, which may serve as a therapeutic target for treatment of autosomal dominant hyper IgE syndrome and other immune disorders.
Metformin: A Potential Therapeutic Tool for Rheumatologists.
Teresa Salvatore,Pia Clara Pafundi,Raffaele Galiero,Klodian Gjeloshi,Francesco Masini,Carlo Acierno,Anna Di Martino,Gaetana Albanese,Maria Rosaria Alfano,Luca Rinaldi,Ferdinando Carlo Sasso +10 more
TL;DR: A large proportion of rheumatologic patients can currently benefit from metformin, such as those with concomitant obesity and type 2 diabetes, two conditions strongly associated with r heumatoid arthritis, osteoarthritis, and gout, as well asThose with diabetes secondary to steroid therapy.
Follicular helper T cells: potential therapeutic targets in rheumatoid arthritis.
TL;DR: In this article, a review summarizes current knowledge of the involvement of Follicular helper T (Tfh) cells in RA pathogenesis and highlights the potential of these cells as therapeutic targets.
T-helper-2 cells and atopic disease: lessons learnt from inborn errors of immunity.
TL;DR: Inborn errors of immunity (IEI) are caused by monogenic variants that affect the host response to bacterial, viral, and fungal pathogens as mentioned in this paper , resulting in severe, recurrent, and life-threatening infections.
Inherited human ZNF341 deficiency.
Vivien Béziat,Claire Fieschi,Mana Momenilandi,Mélanie Migaud,Brahim Belaid,Reda Djidjik,Anne Puel +6 more
TL;DR: A review of the role of ZNF341 in the development of hyper-IgE syndromes can be found in this article , where the authors discuss possible roles for this protein in addition to its control of STAT3 activity.

Bcl6 and Blimp-1 are reciprocal and antagonistic regulators of T follicular helper cell differentiation.
Robert J. Johnston,Amanda C. Poholek,Daniel DiToro,Isharat Yusuf,Danelle Eto,Burton E. Barnett,Alexander L. Dent,Joe Craft,Shane Crotty,Shane Crotty +9 more
TL;DR: It is found that expression of the transcription factor Bcl6 in CD4+ T cells is both necessary and sufficient for in vivo TFH differentiation and T cell help to B cells in mice, and that Bcl 6 and Blimp-1 play central but opposing roles inTFH differentiation.
Bcl6 Mediates the Development of T Follicular Helper Cells
Roza Nurieva,Yeonseok Chung,Gustavo J. Martinez,Xuexian O. Yang,Shinya Tanaka,Tatyana D. Matskevitch,Yi Hong Wang,Chen Dong +7 more
TL;DR: It is demonstrated that the transcription factor Bcl6 is both necessary and sufficient for TFH differentiation and subsequent B cell–mediated immunity, suggesting that it is a master regulator of this lineage.

Follicular B Helper T Cells Express Cxc Chemokine Receptor 5, Localize to B Cell Follicles, and Support Immunoglobulin Production
Dagmar Breitfeld,Lars Ohl,Elisabeth Kremmer,Joachim W. Ellwart,Federica Sallusto,Martin Lipp,Reinhold Förster +6 more
TL;DR: In this article, expression of CXC chemokine receptor 5 (CXCR5) defines a novel subpopulation of B helper T cells localizing to follicles, based on the characteristic localization within secondary lymphoid organs.

Hannum,Daniel M. Gorman,Donna Rennick,Robert A. Kastelein,Rene de Waal Malefyt,Kevin W. Moore +20 more
TL;DR: A novel member of the hemopoietin receptor family is identified as a subunit of the receptor for IL-23, “IL-23R,” which pairs with IL-12Rβ1 to confer IL- 23 responsiveness on cells expressing both subunits.
