Abstract: Coot is a molecular-graphics application for model building and validation of biological macromolecules. The program displays electron-density maps and atomic models and allows model manipulations such as idealization, real-space refinement, manual rotation/translation, rigid-body fitting, ligand search, solvation, mutations, rotamers and Ramachandran idealization. Furthermore, tools are provided for model validation as well as interfaces to external programs for refinement, validation and graphics. The software is designed to be easy to learn for novice users, which is achieved by ensuring that tools for common tasks are `discoverable' through familiar user-interface elements (menus and toolbars) or by intuitive behaviour (mouse controls). Recent developments have focused on providing tools for expert users, with customisable key bindings, extensions and an extensive scripting interface. The software is under rapid development, but has already achieved very widespread use within the crystallographic community. The current state of the software is presented, with a description of the facilities available and of some of the underlying methods employed.

TL;DR: A crystal structure of human KOP in complex with the potent epoxymorphinan opioid agonist MP1104 and an active-state-stabilizing nanobody is provided and key residues that propagate larger-scale structural rearrangements and transducer binding are illuminated that elucidate the structural determinants of KOP pharmacology, function, and biased signaling.

De novo protein design by deep network hallucination
Ivan Anishchenko,Tamuka M. Chidyausiku,Sergey Ovchinnikov,Samuel J. Pellock,David Baker,David Baker +5 more
TL;DR: Deep networks trained to predict native protein structures from their sequences can be inverted to design new proteins, and such networks and methods should contribute, alongside traditional physically based models, to the de novo design of proteins with new functions.
Near-atomic model of microtubule-tau interactions.
Elizabeth H. Kellogg,Elizabeth H. Kellogg,Nisreen M. A. Hejab,Simon Poepsel,Kenneth H. Downing,Frank DiMaio,Eva Nogales,Eva Nogales +7 more
TL;DR: A near-atomic model of microtubule-bound tau provides an explanation for disease-associated phosphorylation data and leads to a model of tau repeats binding in tandem along protofilaments, tethering together tubulin dimers and stabilizing polymerization interfaces.

Structure and function of Parkin E3 ubiquitin ligase reveals aspects of RING and HECT ligases
B.E. Riley,J.C. Lougheed,K. Callaway,M. Velasquez,E. Brecht,Lan K. Nguyen,Thomas A. Shaler,D. Walker,Y. Yang,K. Regnstrom,L. Diep,Z. Zhang,S. Chiou,Michael P. Bova,D.R. Artis,N.H. Yao,J. Baker,T. Yednock,J.A. Johnston +18 more
TL;DR: It is confirmed that Parkin is a RING/HECT hybrid, and the first crystal structure of an RING-between-RING E3 ligase at atomic resolution is provided, providing insight into this disease-related protein.
Disabling Cas9 by an anti-CRISPR DNA mimic

TL;DR: It is shown that the anti-CRISPR protein AcrIIA4 binds only to assembled Cas9–single-guide RNA (sgRNA) complexes and not to Cas9 protein alone, demonstrating that inhibitors can modulate the extent and outcomes of Cas9-mediated gene editing.
