Exosomes derived from alcohol-treated hepatocytes horizontally transfer liver specific miRNA-122 and sensitize monocytes to LPS
TL;DR: It is shown that the number of exosomes significantly increases in the sera of healthy individuals after alcohol binge drinking and in mice after binge or chronic alcohol consumption, and hepatocyte-derived miRNA-122 can reprogram monocytes inducing sensitization to LPS.

Abstract: Hepatocyte damage and inflammation in monocytes/macrophages are central to the pathogenesis of alcoholic hepatitis (AH). MicroRNAs (miRNAs) regulate all of these processes. MiRNA-122 is abundantly expressed in hepatocytes while monocytes/macrophages have low levels. The role of exosomes in AH and possible cross talk between hepatocyte-derived exosomes and immune cells is not explored yet. Here, we show that the number of exosomes significantly increases in the sera of healthy individuals after alcohol binge drinking and in mice after binge or chronic alcohol consumption. Exosomes isolated from sera after alcohol consumption or from in vitro ethanol-treated hepatocytes contained miRNA-122. Exosomes derived from ethanol-treated Huh7.5 cells were taken up by the recipients THP1 monocytes and horizontally transferred a mature form of liver-specific miRNA-122. In vivo, liver mononuclear cells and Kupffer cells from alcohol-fed mice had increased miRNA-122 levels. In monocytes, miRNA-122 transferred via exosomes inhibited the HO-1 pathway and sensitized to LPS stimulation and increased levels of pro-inflammatory cytokines. Finally, inflammatory effects of exosomes from ethanol-treated hepatocytes were prevented by using RNA interference via exosome-mediated delivery of a miRNA-122 inhibitor. These results demonstrate that first, exosomes mediate communication between hepatocytes and monocytes/macrophages and second, hepatocyte-derived miRNA-122 can reprogram monocytes inducing sensitization to LPS.

TL;DR: The CF2 fraction enriched in exosomal markers was the most active on renal tubular cell proliferation and protection from apoptosis and the results demonstrated heterogeneity in quantity and composition of MSC EVs.
Immunological mechanisms and therapeutic targets of fatty liver diseases
TL;DR: It is pointed out that inflammation also plays important roles in promoting liver repair and controlling bacterial infections, and the complex regulatory process of disrupted homeostasis during the development of fatty liver diseases may lead to the developed of improved targeted therapeutic intervention strategies.
Forecasted 2040 global prevalence of nonalcoholic fatty liver disease using hierarchical bayesian approach
TL;DR: By 2040, over half the adult population is forecasted to have NAFLD, with the largest increases occur in women, smokers and those without metabolic syndrome.
Apoptotic tumor cell-derived microRNA-375 uses CD36 to alter the tumor-associated macrophage phenotype.

TL;DR: It is shown that an alternative mode of transfer whereby miR-375 from apoptotic tumour cells can be transferred to tumour-associated macrophages via CD36 receptor, which induces macrophage migration and infiltration to the tumours.

Extracellular vesicles as biomarkers in liver diseases: a clinician’s point of view
TL;DR: The review shows that extracellular vesicles could be useful tools to evaluate activity and fibrosis in non-alcoholic fatty liver disease, predict risk of hepatitis B virus reactivation, predict complications and mortality in cirrhosis, detect early hepatocellular carcinoma, detect malignant transformation in primary sclerosing cholangitis and predict outcomes in acute liver failure.

Exosome-mediated transfer of mRNAs and microRNAs is a novel mechanism of genetic exchange between cells
TL;DR: It is shown that exosomes contain both mRNA and microRNA, which can be delivered to another cell, and can be functional in this new location, and it is proposed that this RNA is called “exosomal shuttle RNA” (esRNA).

TL;DR: 34 novel miRNAs were identified by tissue-specific cloning of approximately 21-nucleotide RNAs from mouse and a miRNA was identified that appears to be the fruitfly and mammalian ortholog of C. elegans lin-4 stRNA.

TL;DR: It is demonstrated by immunoelectron microscopy that the limiting membrane of MIICs can fuse directly with the plasma membrane, resulting in release from the cells of internal MHC class II-containing vesicles, suggesting a role for exosomes in antigen presentation in vivo.
