Abstract: The present invention relates to methods of immunization of aquaculture species by introducing DNA expression systems into the aquaculture species. Such DNA expression systems preferably include DNA sequences encoding polypeptides of pathogens of species of aquaculture. The present invention also relates to methods of administration of DNA expression systems into aquaculture. Such methods include injection, spray, and immersion techniques. The methods of this invention are useful for prophylactic vaccination or therapeutic immunization of fin-fish, shellfish, or other aquatic animals against infectious diseases.

Direct gene transfer into mouse muscle in vivo.

TL;DR: RNA and DNA expression vectors containing genes for chloramphenicol acetyltransferase, luciferase, and beta-galactosidase were separately injected into mouse skeletal muscle in vivo and expression was comparable to that obtained from fibroblasts transfected in vitro under optimal conditions.

Immunostimulatory DNA Sequences Necessary for Effective Intradermal Gene Immunization
Yukio Sato,Mark Roman,H Tighe,Delphine J. Lee,Maripat Corr,Minh-Duc Nguyen,Gregg J. Silverman,Martin Lotz,Dennis A. Carson,Eyal Raz +9 more
TL;DR: Although ISS are necessary for gene vaccination, they down-regulate gene expression and thus may interfere with gene replacement therapy by inducing proinflammatory cytokines.

Direct gene transfer into skeletal muscle in vivo: factors affecting efficiency of transfer and stability of expression.
TL;DR: It is determined that much of the variability of expression is due to nonuniform distribution of substances injected into skeletal muscle in vivo, and a model to ameliorate this is developed.

In vivo promoter activity and transgene expression in mammalian somatic tissues evaluated by using particle bombardment.
TL;DR: The particle bombardment method of gene transfer provides an alternative approach for analysis of in vivo promoter activity and transgene expression in mouse, rabbit, and rhesus monkey tissues and suggests that particle bombardment provides an effective system for studies of in vitro gene transfer and gene therapy.

Plasmid DNA Is Superior to Viral Vectors for Direct Gene Transfer into Adult Mouse Skeletal Muscle
TL;DR: The surprisingly high relative efficiency of pure plasmid DNA suggests that this method will provide a simple, safe and viable alternative for gene therapy involving muscle tissue.
