Abstract: After nearly 10 years of PCR-based analysis of prokaryotic small-subunit ribosomal RNAs for ecological studies it seems necessary to summarize reported pitfalls of this approach which will most likely lead to an erroneous description on the microbial diversity of a given habitat. The following article will cover specific aspects of sample collection, cell lysis, nucleic acid extraction, PCR amplification, separation of amplified DNA, application of nucleic probes and data analysis.

Impact of the Cultivated-Land-Management Scale on Fertilizer Reduction—Empirical Evidence from the Countryside of China
Wenhao Song,Chunhui He +1 more
TL;DR: Wang et al. as mentioned in this paper evaluated the impact of cultivated land management scale on fertilizer reduction via the OLS regression method and found that the fragmentation of cultivated fields is a barrier to decreasing fertilizer application.
Changes in bacterial community composition along a solar radiation gradient in humic waters
TL;DR: Vertical changes in the production of dissolved inorganic carbon, oxygen consumption and bacterial production coincided with changes in BCC, suggesting a relationship between bacterial community composition and function.

Challenges in determining causation in structure–function studies using molecular biological techniques
TL;DR: Various causation criteria improving on the Koch–Henle postulates are presented and approaches for determining cause–effect relationships are illustrated using 16S rDNA-based investigations of filaments that cause bulking and foaming in activated sludge.

Profiling 16S bacterial DNA and RNA: Difference between community structure and transcriptional activity in phenanthrene polluted sand in the vicinity of plant roots
TL;DR: It is concluded that depending on distance from roots, the rhizosphere selected different bacterial communities, and different active species within these communities, resulting in different degradation values.

Phylogenetic identification and in situ detection of individual microbial cells without cultivation.
TL;DR: Phylogenetic analysis of the retrieved rRNA sequence of an uncultured microorganism reveals its closest culturable relatives and may, together with information on the physicochemical conditions of its natural habitat, facilitate more directed cultivation attempts.
