Crossing the Vascular Wall: Common and Unique Mechanisms Exploited by Different Leukocyte Subsets during Extravasation
TL;DR: Current knowledge on regulatory mechanisms of leukocyte extravasation from a leukocytes and endothelial point of view are summarized and common and unique mechanisms that specific leuk cell subsets exploit to succeed in crossing endothelial monolayers are highlighted.

Abstract: Leukocyte extravasation is one of the essential and first steps during the initiation of inflammation. Therefore, a better understanding of the key molecules that regulate this process may help to develop novel therapeutics for treatment of inflammation-based diseases such as atherosclerosis or rheumatoid arthritis. The endothelial adhesion molecules ICAM-1 and VCAM-1 are known as the central mediators of leukocyte adhesion to and transmigration across the endothelium. Engagement of these molecules by their leukocyte integrin receptors initiates the activation of several signaling pathways within both leukocytes and endothelium. Several of such events have been described to occur during transendothelial migration of all leukocyte subsets, whereas other mechanisms are known only for a single leukocyte subset. Here, we summarize current knowledge on regulatory mechanisms of leukocyte extravasation from a leukocyte and endothelial point of view, respectively. Specifically, we will focus on highlighting common and unique mechanisms that specific leukocyte subsets exploit to succeed in crossing endothelial monolayers.

HS1 deficiency impairs neutrophil recruitment in vivo and activation of the small GTPases Rac1 and Rap1
Joanna Latasiewicz,Annette Artz,Ding Jing,Mariana Pacheco Blanco,Silke Maria Currie,Martha Velázquez Avila,Michael Schnoor,Dietmar Vestweber +7 more
TL;DR: HS1 is established as an important regulator of proper Rac1 and Rap1 activation and neutrophil extravasation in vivo and with the use of Rac1 KO neutrophils, it is shown that Rac1 acts upstream of Rap1.

Soluble DC-HIL/Gpnmb Modulates T-lymphocyte Extravasation to Inflamed Skin.
TL;DR: In this paper, the effects of sDC-HIL binding to EC on T-cell extravasation using an allergic contact dermatitis model in mice were examined, and it was shown that the crosstalk between T-cells and EC mediates cross-talk between the two pathways, regulating the cutaneous immune response.

Altered DNA methylation in a severe subtype of idiopathic autism: Evidence for sex differences in affected metabolic pathways.
TL;DR: It is shown for the first time that DNA methylation differences between autistic and sex-matched non-autistic siblings are in part sex dependent, with differentially methylated genes in females associated with pathways that implicate mitochondrial dysfunction and metabolic disorders that may offer some protection to females against autism spectrum disorders.

Intermittent rolling is a defect of the extravasation cascade caused by Myosin1e-deficiency in neutrophils.
Eduardo Vadillo,Sandra Chánez-Paredes,Hilda Vargas-Robles,Idaira M. Guerrero-Fonseca,Ramón Castellanos-Martínez,Alexander García-Ponce,Porfirio Nava,Daniel Alberto Girón-Pérez,Leopoldo Santos-Argumedo,Michael Schnoor +9 more
TL;DR: It is shown that absence of myosin1e inhibits neutrophil migration out of the blood stream into inflamed tissue, and a striking discontinuous rolling behavior termed “intermittent rolling,” during which Myo1e-deficient neutrophils showed alternating rolling and jumping movements, could be a therapeutic target for chronic inflammatory diseases.

Megakaryocyte volume modulates bone marrow niche properties and cell migration dynamics
Maximilian G. Gorelashvili,Oguzhan Angay,Katherina Hemmen,Vanessa Klaus,David Stegner,Katrin G. Heinze +5 more
TL;DR: It is found that megakaryocytes influence migration of neutrophils and hematopoietic stem cells, and thus act as biomechanical restrainers modulating cell mobility and extravasation in platelet-depleted mice and neutrophil mobility was reduced in these animals.

Two types of murine helper T cell clone. I. Definition according to profiles of lymphokine activities and secreted proteins.
TL;DR: A panel of antigen-specific mouse helper T cell clones was characterized according to patterns of lymphokine activity production, and two types of T cell were distinguished.

Chemokines and leukocyte traffic
TL;DR: Over the past ten years, numerous chemokines have been identified as attractants of different types of blood leukocytes to sites of infection and inflammation and are now known to also function as regulatory molecules in leukocyte maturation, traffic and homing of lymphocytes, and the development of lymphoid tissues.
