Soft lithography for microfluidics: a review
20 Mar 2008 - Biochip Journal

Abstract:

Soft lithography has provided a low-expertise route toward micro/nanofabrication and is playing an important role in microfluidics, ranging from simple channel fabrication to the creation of micropatterns onto a surface or within a microfluidic channel. Soft lithography has provided a low-expertise route toward micro/nanofabrication and is playing an important role in microfluidics, ranging from simple channel fabrication to the creation of micropatterns onto a surface or within a microfluidic channel. In this review, the materials, methods, and applications of soft lithography for microfluidics are briefly summarized with a particular emphasis on integrated microfluidic systems containing physical microstructures or a topographically patterned substrate. Relevant exemplary works based on the combination of various soft lithographic methods using microfluidics are introduced with some comments on their merits and weaknesses. read more read less

As a glucose biosensing principle, a colorimetric glucose assay, using glucose oxidase (GOx) and horseradish peroxidase (HRP), was chosen. The enzymatic glucose assay was implanted on the analytical paper-based device, which is fabricated by the wax printing method. The fabricated device consists of two paper layers. The top layer has a sample loading zone and a detection zone, which are modified with enzymes and chromogens. The bottom layer contains a fluidic channel to convey the solution from the loading zone to the detection zone. Double-sided adhesive tape is used to attach these two layers. In this system, when a glucose solution is dropped onto the loading zone, the solution is transferred to the detection zone, which is modified with GOx, HRP, and chromogenic compounds through the connected fluidic channel. In the presence of GOx-generated H2O2, HRP converts chromogenic compounds into the final product exhibiting a blue color, inducing color change in the detection zone. To confirm the changes in signal intensity in the detection zone, the resulting image was registered by a digital camera from a smartphone. To minimize signal interference from external light, the experiment was performed in a specifically designed light-tight box, which was suited to the smartphone. By using the developed biosensing system, various concentrations of glucose samples (0–20 mM) and human serum (5–17 mM) were precisely analyzed within a few minutes. With the developed system, we could expand the applicability of a smartphone to bioanalytical health care. read more read less

Abstract Measuring morphological and biochemical features of tissue is crucial for disease diagnosis and surgical guidance, providing clinically significant information related to pathophysiology. Hyperspectral imaging (HSI) techniques obtain both spatial and spectral features of tissue without labeling molecules such as fluorescent dyes, which provides rich information for improved disease diagnosis and treatment. Recent advances in HSI systems have demonstrated its potential for clinical applications, especially in disease diagnosis and image-guided surgery. This review summarizes the basic principle of HSI and optical systems, deep-learning-based image analysis, and clinical applications of HSI to provide insight into this rapidly growing field of research. In addition, the challenges facing the clinical implementation of HSI techniques are discussed. read more read less

The architecture of biofilm plays an important role in protecting bacteria under physical, chemical, and biological stress. The shear stress is one of the major factors to construct stable bioflim. The experimental observation of biofilm formation on large-scale water flow has been limited because most of fluid pipe are water and sewer lines. This study presents the biofilm formation in a PDMS-based microfluidic channel which is able to simulate fluid pipes at small scale. We could characterize the hydrodynamics of the growth of single-species bacteria between biofilm formation and the external environmental factors. Particularly, the dynamics of biofilm formation confirms that biofilm, under the optimum shear stress, efficiently form a stable EPS structure which provides a mechanical shield against high-pressure fluidic flow. read more read less
