Current analytical methods for plant auxin quantification – A review

Accurate quantification of phytohormones in specific target cells will greatly facilitate the elucidation of the molecular mechanisms of their actions. Since the Avena coleoptile curvature test for auxin was established in the 1920s, phytohormonal assays have undergone a long march involving several technological generations. Biological methods such as traditional bioassays and immunoassays dominated phytohormonal quantification from the 1920s to the 1980s. In the past three decades, physical methods based on chromatography and mass spectrometry have largely replaced biological methods and have become the mainstream methods for phytohormonal quantification. However, even the most advanced tandem mass spectrometry today is still unable to perform quantification or to reveal hormone dynamics at the single-cell level. Some molecular phytohormonal quantification methods such as phytohormone-inducible promoter-reporter systems and degrons (phytohormone-inducible signaling repressor protein degradation) have shown the potential for single-cell-level quantification. Furthermore, biosensors offer the ability for fast determination while “nucleic acid terminal protection” methods provide the possibility for improvement of sensitivity. Therefore, phytohormonal quantification based on biological principles has regained the attention from plant biologists and will become increasingly important in future phytohormonal quantification practice. In this chapter, phytohormonal quantification based on biological principles, as well as the sample preparation, localization, and profiling techniques have been reviewed. Moreover, perspectives of ongoing approaches with single-cell dynamics are also discussed.

Phytohormonal quantification based on biological principles

In this paper, a novel flow-injection chemiluminescence method was developed for the determination of indole-3-acetic acid (IAA). The chemiluminescence signal from the reaction of an Ag(III) complex and a sulfuric acid system was enhanced in the presence of IAA. The conditions of the CL system were investigated and optimized. Under the optimal conditions, the relative chemiluminescence intensity was linear with the IAA concentration in the range of 1.0 × 10−10 g mL−1 to 1 × 10−8 g mL−1. The detection limit for IAA was 7.7 × 10−11 g mL−1, and the relative standard deviation (n = 11) was 0.5%. The proposed method was applied to the analysis of IAA in human urine, mung bean sprouts and soil samples with recoveries of 103.5–117.1%, 87.0–98.4% and 94.1–118.8%, respectively, and the relative standard deviations was 0.6–2.7%. The results obtained by the proposed method agreed well with those obtained from the HPLC method. The chemiluminescence mechanism is discussed by comparison of fluorescence spectra and the UV-vis absorption spectra.

A novel flow injection chemiluminescence method for the determination of indole-3-acetic acid in biological samples by using trivalent silver

A sensitive simultaneous electroanalysis of phytohormones indole-3-acetic acid (IAA) and salicylic acid (SA) based on a novel copper nanoparticles-chitosan film-carbon nanoparticles-multiwalled carbon nanotubes (CuNPs-CSF-CNPs-MWCNTs) composite was reported. CNPs were prepared by hydrothermal reaction of chitosan. Then the CuNPs-CSF-CNPs-MWCNTs composite was facilely prepared by one-step co-electrodeposition of CuNPs and CNPs fixed chitosan residues on modified electrode. Scanning electron microscope (SEM), transmission electron microscopy (TEM), selected area electron diffraction (SAED), energy dispersive spectroscopy (EDS), X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FT-IR), cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS), and linear sweep voltammetry (LSV) were used to characterize the properties of the composite. Under optimal conditions, the composite modified electrode had a good linear relationship with IAA in the range of 0.01–50 μM, and a good linear relationship with SA in the range of 4–30 μM. The detection limits were 0.0086 μM and 0.7 μM (S/N = 3), respectively. In addition, the sensor could also be used for the simultaneous detection of IAA and SA in real leaf samples with satisfactory recovery.

One-Step Co-Electrodeposition of Copper Nanoparticles-Chitosan Film-Carbon Nanoparticles-Multiwalled Carbon Nanotubes Composite for Electroanalysis of Indole-3-Acetic Acid and Salicylic Acid

A surface plasmon resonance (SPR)-based detection methodology, which allows the identification and quantification of the plant hormone, 1H-indole-3-acetic acid (IAA) in Peach, Rosa flower, and Crape myrtle bud is described. To achieve high selectivity, a molecularly imprinted monolayer (MIM) was prepared from alkanethiols self-assembled on an SPR sensor chip with preadsorbed template (IAA). The association constant of the MIM to IAA was calculated to be 2.8 × 1011 M−1 by analysis of the SPR wavenumber changes induced by different concentrations of IAA. A good calibration curve was obtained in a low concentration range of IAA. Moreover, the MIM revealed impressive selectivity with the selectivity factors of 1.0, 0.12, and 0.20 for IAA and other related analogues.
