Microbial electrochemical bioremediation of petroleum hydrocarbons (PHCs) pollution: Recent advances and outlook

Distinct microbiota assembly mechanisms revealed in different reconstruction stages during gut regeneration in the sea cucumber Apostichopus japonicus

Trillions of commensal bacteria reside in the digestive tract of animals and constitute a multifunctional system called the gut microbiota. As an integral part of an animal organ, it coevolved with the host to constitute a complex and symbiotic relationship. It is increasingly recognized that the gut microbiota plays crucial roles in many biological processes, including nutrient digestion, energy metabolism and storage, immune system modulation, neurotransmission, and even behavior regulation. In this chapter, we will address the composition and function of the gut bacterial communities of sea cucumbers in different habitats, the gut microbiota of sea cucumbers during distinctive life processes (such as regeneration), and various endogenous and exogenous factors that can affect it. The aim is to explore the source, dynamic change, and function of intestinal bacteria in sea cucumbers and uncover the relationship between the gut microbiota and the health and unique habits of its host. 

Gut microbiota of sea cucumbers, with a focus on Apostichopus japonicus

During the seedlings culture of sea cucumbers including Holthuria leucospilota, natural microalgae are generally used as ideal food for the pelagic larvae of sea cucumbers, which lead to a strong demand for microalgae production. However, microalgae production is time-consuming and laborious and susceptible to pests and variable whether, and unstable supply of microalgae often results in culture failure of sea cucumber larvae. In this study, we tested the effect of three different diets on the growth, survival rate, and internal microbiota composition of sea cucumbers aiming to develop an artificial diet for H. leucospilota larvae. The results showed that the sea cucumber larvae fed with the mixed artificial diet (composed of Sargassum powder, chilled macroalgae solution, wall-breaking Chlorella powder, and yeast powder) had the most balanced growth rate, highest survival rate, and a higher α-diversity and abundance of Plesiocystis, Haloferula, and Firmicutes in body when they were compared to the larvae fed with the natural bait, Chaetoceros muelleri. However, the abundance of potential pathogenic bacteria, Francisella and Acinetobacter, was higher in the larvae fed with the natural bait. In conclusion, the mixed artificial diet was developed for H. leucospilota larval culture, which can be used as a good alternative to live microalga with cumbersome processes. 

Developing artificial mixed diets for larval culture of sea cucumber, Holthuria leucospilota, and their effects on the internal microbiota

Tetrabromobisphenol A (TBBPA), a commonly utilized brominated flame retardant, is found in many types of abiotic and biotic matrices. TBBPA can increase oxidative stress, disrupt the endocrine system, cause neurodevelopmental disorders and activate peroxisome proliferator-activated receptors to modulate lipid deposits in aquatic animals. However, the toxic mechanism of TBBPA on the gut microbiota and intestinal health remains unclear. Apostichopus japonicus is an ideal model for studying the relationship between environmental contaminants and intestinal health due to its unique capacity for evisceration and quickly regenerated intestine. In the present study, we investigated the toxic mechanism of TBBPA on the gut microbiota and intestinal health in the regenerated intestine of A. japonicus. The results show that TBBPA exposure decreased the health of the regenerated intestine and the enzymatic activities, alpha diversity indices, and the relative abundance of the gut microbiota. Transcriptome analysis shows that TBBPA exposure affected lipid metabolism via the PPAR signaling pathway during the process of intestinal regeneration in A. japonicus, suggesting that TBBPA exposure can affect the composition and function of the gut microbiota and intestinal health in the regenerated intestine of A. japonicus. These results provide a basis for further research on the potential toxicity of TBBPA to the intestinal health in animals.
