Engineered wound dressing materials with excellent injectability, self-healing ability, tissue-adhesiveness, especially the ones possessing potential therapeutic effects have great practical significance in healthcare. Herein, an injectable quaternary ammonium chitosan (QCS)/tannic acid (TA) hydrogel based on QCS and TA was designed and fabricated by facile mixing of the two ingredients under physiological conditions. In this system, hydrogels were mainly cross-linked by dynamic ionic bonds and hydrogen bonds between QCS and TA, which endows the hydrogel with excellent injectable, self-healing, and adhesive properties. Benefitting from the inherent antioxidative, antibacterial, and hemostatic abilities of TA and QCS, this hydrogel showed superior reactive oxygen species scavenging activity, broad-spectrum antibacterial ability, as well as rapid hemostatic capability. Moreover, the QCS/TA2.5 hydrogel (containing 2.5% TA) exhibited excellent biocompatibility. The in vivo experiments also showed that QCS/TA2.5 hydrogel dressing not only rapidly stopped the bleeding of arterial and deep incompressible wounds in mouse tail amputation, femoral artery hemorrhage, and liver incision models but also significantly accelerated wound healing in a full-thickness skin wound model. For the great potentials listed above, this multifunctional QCS/TA2.5 hydrogel offers a promising network as a dressing material for both rapid hemostasis and skin wound repair.

Abstract Microbial infections based on drug‐resistant pathogenic organisms following surgery or trauma and uncontrolled bleeding are the main causes of increased mortality from trauma worldwide. The prevalence of drug‐resistant pathogens has led to a significant increase in medical costs and poses a great threat to the normal life of people. This is an important issue in the field of biomedicine, and the emergence of new antimicrobial materials hydrogels holds great promise for solving this problem. Hydrogel is an important material with good biocompatibility, water absorption, oxygen permeability, adhesion, degradation, self‐healing, corrosion resistance, and controlled release of drugs as well as structural diversity. Bacteria‐disturbing hydrogels have important applications in the direction of surgical treatment, wound dressing, medical device coating, and tissue engineering. This paper reviews the classification of antimicrobial hydrogels, the current status of research, and the potential of antimicrobial hydrogels for one application in biomedicine, and analyzes the current research of hydrogels in biomedical applications from five aspects: metal‐loaded hydrogels, drug‐loaded hydrogels, carbon‐material‐loaded hydrogels, hydrogels with fixed antimicrobial activity and biological antimicrobial hydrogels, and provides an outlook on the high antimicrobial activity, biodegradability, biocompatibility, injectability, clinical applicability and future development prospects of hydrogels in this field. 

Antimicrobial Hydrogels: Potential Materials for Medical Application.

According to International Diabetes Federation Diabetes Atlas statistics, diabetic retinopathy (DR) is the leading cause of vision loss in blinding diseases. The underlying cause of retinal vasculopathy progression in diabetic patients is hyperglycemia and hypoxia features in microvascular region. Hence, cyanobacteria are used as carriers to load both gold nanoparticles (Au NPs) with glucose oxidase-like activity and iridium nanoparticles (Ir NPs) with catalase-like activity, respectively ([email protected]@Ir). The Au NPs nanozyme first degrades glucose into hydrogen peroxide, which is further decomposed into H2O and O2 by the Ir NPs to complete the cascade hypoglycemic reaction. Based on the unique light transmittance of eyeball and the accumulation of light in the retinal area, the sustainable O2 production by Cyano greatly alleviates the hypoxia of microenvironment, leading to the decrease of angiogenic growth factor and hypoxia-inducible factor expressions. Simultaneously, the highly expressed peroxide in the DR microenvironment can also be eliminated by Ir NPs for anti-inflammatory property. Furthermore, it is demonstrated in DR animal model that [email protected]@Ir significantly reduces neovascular progression and vascular leakage. This novel treatment mode fundamentally degrades blood glucose, continuously supplies O2, and scavenges free radicals for comprehensive microenvironment regulation, providing inspirations for solving fundus complications of DR. 

A Novel Photosynthetic Biohybrid System for Microenvironment Regulation of Diabetes Retinopathy through Continuous Oxygen Supply and Nanozyme Cascade Reaction

Corneal neovascularization (CoNV)-induced blindness is an enduring and challenging condition with limited management options. Small interfering RNA (siRNA) is a promising strategy for preventing CoNV. This study reported a new strategy using siVEGFA to silence vascular endothelial growth factor A (VEGFA) for CoNV treatment. To improve the efficacy of siVEGFA delivery, a pH-sensitive polycationic mPEG2k-PAMA30-P(DEA29-D5A29) (TPPA) was fabricated. TPPA/siVEGFA polyplexes enter cells via clathrin-mediated endocytosis, resulting in higher cellular uptake efficiency and comparable silencing efficiency than that of Lipofectamine 2000 in vitro. Hemolytic assays verified that TPPA safe in normal physiological environments (pH 7.4) but can easily destroy membranes in acidic mature endosomes (pH 4.0). Studies on the distribution of TPPA in vivo showed that it could prolong the retention time of siVEGFA and promote its penetration in the cornea. In a mouse model induced by alkali burn, TPPA efficiently delivered siVEGFA to the lesion site and achieved VEGFA silencing efficiency. Importantly, the inhibitory effect of TPPA/siVEGFA on CoNV was comparable to that of the anti-VEGF drug ranibizumab. Delivering siRNA using pH-sensitive polycations to the ocular environment provides a new strategy to efficiently inhibit CoNV.

Efficient ocular delivery of siRNA via pH-sensitive vehicles for corneal neovascularization inhibition

Posterior capsular opacification (PCO), the highest incidence complication after cataract surgery, is mainly due to the attachment, proliferation, and migration of the residual lens epithelial cells (LECs). Although the drug-eluting IOLs have been proved to be an effective way to prevent PCO incidence, its preparations are time consuming and require tedious preparation steps. Herein, the thermoreversible agarose is adopted to prepare drug-eluting IOL. Such functional coating can be obtained easily by simple immersion in the antiproliferative drug containing hot agarose and taken out for cooling, which not only does not affect the optical property but also can effectively decrease the PCO incidence after intraocular implantation. As a result, the proposed agarose coating provides a rapid and economical alternative of drug-eluting IOL fabrication for PCO prevention.
