Gut Microbiome Alterations Precede Cerebral Amyloidosis and Microglial Pathology in a Mouse Model of Alzheimer's Disease.
Yijing Chen,Lihua Fang,Shuo Chen,Haokui Zhou,Yingying Fan,Li Lin,Jing Li,Jinying Xu,Yuewen Chen,Yuewen Chen,Yingfei Ma,Yu Chen,Yu Chen +12 more
TL;DR: Gut microbiota alterations precede the development of key pathological features of AD, including amyloidosis and plaque-localized neuroinflammation, and the investigation of gut microbiota might provide new avenues for developing diagnostic biomarkers and therapeutic targets for AD.
Identification of genetic risk factors in the Chinese population implicates a role of immune system in Alzheimer's disease pathogenesis
Xiaopu Zhou,Yu Chen,Yu Chen,Kin Y. Mok,Kin Y. Mok,Qianhua Zhao,Keliang Chen,Yuewen Chen,Yuewen Chen,John Hardy,Yun Li,Amy K.Y. Fu,Qihao Guo,Nancy Y. Ip +13 more
TL;DR: Network analysis of hippocampus and blood transcriptome datasets suggests that the risk variants in the APOE, GCH1, and KCNJ15 loci might exert their functions through their regulatory effects on immune-related pathways during AD pathogenesis.

Brain-wide Cas9-mediated cleavage of a gene causing familial Alzheimer’s disease alleviates amyloid-related pathologies in mice
Yangyang Duan,Tao Ye,Zhe Qu,Yuewen Chen,Yuewen Chen,Abigail Miranda,Xiaopu Zhou,Ka-Chun Lok,Yu Chen,Amy K.Y. Fu,Viviana Gradinaru,Nancy Y. Ip +11 more
TL;DR: In this paper, the brain-wide selective disruption of a mutated APP allele in transgenic mouse models carrying the human APP Swedish mutation alleviates amyloid-beta-associated pathologies for at least six months after a single intrahippocampal administration of an adeno-associated virus that encodes both Cas9 and a single-guide RNA that targets the mutation.

Coronin 2B regulates dendrite outgrowth by modulating actin dynamics
Yuewen Chen,Yuewen Chen,Jinying Xu,Yulin Zhang,Shuangshuang Ma,Wanying Yi,Saijuan Liu,Xiaojun Yu,Jiali Wang,Yu Chen,Yu Chen +10 more
TL;DR: It is suggested that coronin 2B is important for promoting dendrite outgrowth by limiting the speed of actin polymerization at growth cones.

Efficient manipulation of gene dosage in human iPSCs using CRISPR/Cas9 nickases.
Tao Ye,Yangyang Duan,Hayley W. S. Tsang,He Xu,Yuewen Chen,Yuewen Chen,Han Cao,Yu Chen,Yu Chen,Amy K.Y. Fu,Nancy Y. Ip +10 more
- 12 Feb 2021
TL;DR: In this paper, the authors demonstrate a simple and precise method to simultaneously generate iPSC lines with different gene dosages using paired Cas9 nickases, which enables the rapid and simultaneous generation of iPSCs with monoallelic, biallelic, or triallelic knockout of APP.
