Evaluation of MALDI-TOF MS, sequencing of D2 LSU rRNA and internal transcribed spacer regions (ITS) for the identification of filamentous fungi isolated from a pharmaceutical facility.

This article is the third in the Asian Journal of Mycology Notes series, wherein we report 50 new fungal collections distributed in two phyla, five classes, 16 orders and 35 families. The present study provides descriptions and illustrations for five new species (Acrocalymma hyaline, Allocryptovalsa aquilariae, Alternaria arida, Apoharknessia thailandica and Tatraea aseptata) and 21 new host and 23 new geographical records. All these introductions are supported by morphological data and multi-gene phylogenetic analyses. This article provides the platform to disseminate the data on fungal collections with new sequence data, which is vital for future studies. An accurate and timely report of new fungus-host or fungus-country records is necessary for the diagnostics, identification, and management of economically significant fungal groups, especially the phytopathogens.

The Sugar Maple (Acer saccharum) is a keystone species in Eastern North America. Despite this, taxonomic surveys enumerating the fungal populations of the bark of this tree species are lacking. Using traditional (direct observation and moist chambers) and high-throughput (particle filtration with dilution to extinction) culturing methods, this study examines the microfungi observed and isolated from A. saccharum bark and compares the efficiency of the different isolation methods employed. Bark collected from five different dead trees, from three distinct forests provide an insight on the feasibility of the high-throughput method in isolating microfungi from this substrate, an approach never previously experimented from A. saccharum bark. Commonly isolated taxa are discussed and compared to prior studies. Three novel species, which were commonly isolated, are described and compared to closely related taxa based on morphological and concatenated multi-gene phylogenetic analyses. The Biodiversity of Microfungi Isolated from the Bark of the Sugar Maple (Acer saccharum)

This article presents the results of an ongoing inventory of Ascomycota in Yunnan, China, carried out as part of the research project series “Exploring ascomycete diversity in Yunnan”. From over 100 samples collected from diverse host substrates, microfungi have been isolated, identified and are currently being documented. The primary objective of this research is to promote the discovery of novel taxa and explore the ascomycete diversity in the region, utilising a morphology-phylogeny approach. This article represents the second series of species descriptions for the project and introduces three undocumented species found in the families Bambusicolaceae, Dictyosporiaceae and Periconiaceae, belonging to the suborder Massarineae (Pleosporales, Dothideomycetes). These novel taxa exhibit typical morphological characteristics of Bambusicola, Periconia and Trichobotrys, leading to their designation as Bambusicola hongheensis, Periconia kunmingensis and Trichobotrys sinensis. Comprehensive multigene phylogenetic analyses were conducted to validate the novelty of these species. The results revealed well-defined clades that are clearly distinct from other related species, providing robust support for their placement within their respective families. Notably, this study unveils the phylogenetic affinity of Trichobotrys within Dictyosporiaceae for the first time. Additionally, the synanamorphism for the genus Trichobotrys is also reported for the first time. Detailed descriptions, illustrations and updated phylogenies of the novel species are provided, and thus presenting a valuable resource for researchers and mycologists interested in the diversity of ascomycetes in Yunnan. By enhancing our understanding of the Ascomycota diversity in this region, this research contributes to the broader field of fungal taxonomy and their phylogenetic understanding.

The genus Dlhawksworthia presently includes three species. All the previously described species have been isolated from plants. Besides, none of these three species have ever been recorded in China. We conducted surveys in various regions of China to isolate and identify endolichenic fungi associated with diverse lichen species. During these surveys, we isolated both previously known and undescribed fungi associated with lichens. Among these, there was an isolate of an unknown fungus. The morphological and molecular analyses indicated that this isolate represented a new species from the genus Dlhawksworthia. As a consequence, we described this fungus as Dlhawksworthia flavoparmeliae sp. nov. This is the first report of Dlhawksworthia isolated from a lichen in China and globally.

Lichens are the result of a symbiotic interaction between fungi (mycobionts) and algae (phycobionts). Aside from mycobionts, lichen thalli can also contain non-lichenised fungal species, such as lichenicolous and endolichenic fungi. For this study, three surveys were conducted in China’s Yunnan Province and Inner Mongolia Autonomous Region between 2017 and 2020. Several samples of four lichen species were collected during these surveys: Candelariafibrosa, Flavoparmeliacaperata, Flavopuncteliaflaventior and Ramalinasinensis. Six isolates of Coniochaeta were recovered from these four lichen species. The phylogenetic and morphological analyses revealed that two of these isolates were previously identified species, Coniochaetavelutinosa and C.acaciae. Those remaining were from potentially unknown species. We used molecular and morphological data to describe these previously-unknown species as Coniochaetafibrosaesp. nov., C.mongoliaesp. nov. and C.sinensissp. nov. The findings of this study significantly improve our understanding of the variety and habitat preferences of Coniochaeta in China and globally.
