The lysosomes of cancer cells have lower pH and higher viscosity than those of normal cells. These features can be used as sensitive and selective markers for cancer diagnosis. In this work, a pH and viscosity dual responsive lysosome targeting fluorescent probe 1 was designed based on the transformation of amino- and imino- forms of pyronine and the twisted intramolecular charge shuttle (TICS) sensing mechanism. Live cancer cells and tumors were effectively distinguished from normal cells and organs through fluorescence imaging of probe 1, which indicated that probe 1 could serve as an effective tool for visualization of tumors at organ level with high selectivity.

One Stone, Three Birds: pH Triggered Transformation of Aminopyronine and Iminopyronine Based Lysosome Targeting Viscosity Probe for Cancer Visualization.

Understanding the molecular machinery regulating ferroptosis and leveraging this accumulating knowledge could facilitate disease prevention, diagnosis, treatment, and prognosis. The emerging approaches endowing in situ detection of the major regulators of ferroptosis at the cellular and tissue levels, and in living subjects, provide a multi-scale perspective to study ferroptosis. More importantly, ferroptosis-based applications integrating the rationality of ferroptosis detection with technological platforms have shown great promise in preclinical research. In this review, we first briefly summarized the mechanisms and key regulators involved in ferroptosis. Ferroptosis detection approaches were then presented to delineate their design, mechanisms of action, and applications. Special interest was placed on ferroptosis-based applications integrating multifunctional platforms. We hope that this review is timely to spur research interest in ferroptosis-based disease management.

Ferroptosis Detection: From Approaches to Applications.

Polymethine cyanine dyes, as the most important class of organic near-infrared-II (NIR-II) fluorophores, recently received increasing attention due to their high molar extinction coefficients, intensive fluorescence brightness, and flexible wavelength tunability for fluorescent bioimaging applications. Very recently, remarkable advances have been made in the development of NIR-II polymethine fluorophores with improved optical performance, mainly including tunable fluorescence, improved brightness, improved water solubility and stability. In this review, we summarize the recent research advances in molecular tailoring design strategies of NIR-II polymethine fluorophores, and then emphasize the representative bioimaging and biosensing applications. The potential challenges and perspectives of NIR-II polymethine fluorophores in this emerging field are also discussed. This review may provide guidance and reference for further development of high-performance NIR-II polymethine fluorophores to boost their clinical translation in the future.

The pursuit of polymethine fluorophores with NIR-II emission and high brightness for in vivo applications

Chemosensors based on traditional fluorescent dyes have always contributed to the development of chemical sensor areas. In this review, the rhodamine-based chemosensors’ improvements and applications from 2018 to 2022 are discussed, mainly focusing on cations (metal ions and H+), anions (CN−, F−, etc.), and small bio-functional molecules’ (thiols, amino acids, etc.) detection. Specifically, this review highlights the detection target, detection limit, detection solution system, detection mechanism, and performance of the rhodamine-based sensors. Although these rhodamine-based sensors are well developed, their repeatability and sensitivity still need significant improvement. This review is expected to bring new clues and bright ideas to researchers for further advances in rhodamine-based chemosensors in the future.

As a new form of regulated cell death, ferroptosis is closely related to various diseases. To interpret this biological behavior and monitor related pathological processes, it is necessary to develop appropriate detection strategies and tools. Considering that ferroptosis is featured with remarkable lipid peroxidation of various cell membranes, it is logical to detect membranes' structural and environmental changes for the direct assessment of ferroptosis. For this sake, we designed novel polarity-sensitive fluorescent probes Mem-C1C18 and Mem-C18C18, which have superior plasma membrane anchorage, high brightness, and sensitive responses to environmental polarity by changing their fluorescence lifetimes. Mem-C1C18 with much less tendency to aggregate than Mem-C18C18 outperformed the latter in high resolution fluorescence labeling of artificial vesicle membranes and plasma membranes of live cells. Thus, Mem-C1C18 was selected to monitor plasma membranes damaged along ferroptosis process for the first time, in combination with the technique of fluorescence lifetime imaging (FLIM). After treating HeLa cells with Erastin, a typical ferroptosis inducer, the mean fluorescence lifetime of Mem-C1C18 displayed a considerable increase from 3.00 to 4.93 ns, with a 64% increase (corresponding to the polarity parameter Δf increased from 0.213 to 0.232). Therefore, our idea to utilize a probe to quantitate the changes in polarity of plasma membranes proves to be an effective method in the evaluation of the ferroptosis process.

Nitric oxide (NO) is an important cellular messenger molecule in the cardiovascular, nervous and immune systems. Real-time monitoring of NO activity in specific organelles of live cells is important to understand its biological function. In this work, a nucleus targetable ratiometric NO probe, Hoe-Rh-NO, is developed by linking Hoechst to rhodamine spirolactam. The Hoechst part conducts nucleus targeting and the rhodamine spirolactam senses NO. The two fluorophores constitute a NO switchable FRET system for ratiometric imaging. This newly developed probe Hoe-Rh-NO displays good nucleus targeting ability, high sensitivity and selectivity towards NO, low cytotoxicity and most importantly detects NO through ratiometric fluorescence imaging. By using Hoe-Rh-NO, we confirmed the presence of NO in the nucleus and detected endogenous NO during inflammation in cells and zebrafishes.
