Aberrant H3K4me3 modification of epiblast genes of extraembryonic tissue causes placental defects and implantation failure in mouse IVF embryos.
Dandan Bai,Jin Sun,Chuan Chen,Yanping Jia,Yanhe Li,Kuisheng Liu,Yalin Zhang,Jiqing Yin,Yingdong Liu,Xiaoxiao Han,Jingling Ruan,Xiaochen Kou,Yanhong Zhao,Hong Wang,Zheng Wang,Miaoxin Chen,Xiaoming Teng,Cizhong Jiang,Shaorong Gao,Wenqiang Liu +19 more
TL;DR: In this article , a combined histone modification analysis reveals that aberrant H3K4me3 modification at the Epiblast active promoters results in increased expression of these genes in extraembryonic ectoderm (ExE).

Mettl14-driven senescence-associated secretory phenotype facilitates somatic cell reprogramming
Chenxiang Xi,Jiatong Sun,Xiaocui Xu,You Wu,Xiaochen Kou,Yanhong Zhao,Jiachen Shen,Yu Dong,Kang Chen,Zhongqu Su,Dan Liu,W. Ye,Yingdong Liu,Ran Zhang,Yiliang Xu,Hong Wang,Lujiang Hao,Li Wu,Shaorong Gao +18 more
TL;DR: In this paper , Mettl14, another component of the methyltransferase complex, can significantly enhance the generation of induced pluripotent stem cells (iPSCs) in an m6A-independent manner.

Inhibition of Wnt activity improves peri-implantation development of somatic cell nuclear transfer embryos
Yanhe Li,Caihong Zheng,Yingdong Liu,Jincan He,Qiang Zhang,Yalin Zhang,Xiaochen Kou,Yanhong Zhao,Kuisheng Liu,Dandan Bai,Yanping Jia,Xiaoxiao Han,Yifan Sheng,Jiqing Yin,Hong Wang,Shuai Gao,Wenqiang Liu,Shaorong Gao +17 more
TL;DR: A 3D in vitro culture system for SCNT peri-implantation embryos is developed and it is discovered that persistent Wnt signals block the naïve-to-primed pluripotency transition of epiblasts with aberrant H3K27me3 occupancy, which in turn leads to defects in epiblast transformation events and subsequent implantation failure.

Bilineage embryo-like structure from EPS cells can produce live mice with tetraploid trophectoderm
Kuisheng Liu,Xiaocui Xu,Dandan Bai,Yanhe Li,Yalin Zhang,Yanping Jia,Mingyue Guo,Xiaoxiao Han,Yingdong Liu,Yifan Sheng,Xiaochen Kou,Yanhong Zhao,Jiqing Yin,Sheng Liu,Jiayu Chen,Hong Wang,Yixuan Wang,Wenqiang Liu,Shaorong Gao +18 more
TL;DR: This study reveals that TE improvement is critical for constructing a functional embryo using stem cells in vitro using EPS-blastoids.
