Ascomycetes from karst landscapes of Guizhou Province, China

Species in the Botryosphaeriaceae are common plant pathogens, endophytes, and saprobes found on a variety of mainly woody hosts. Botryosphaeriaceae is a high-profile fungal family whose genera have been subjected to continuous revisions in recent years. Surveys conducted during 2019 and 2020 on several decaying woody hosts (from dead arial twigs, branches, stems, bark, and seed pods) in China and Thailand revealed a high diversity of Botryosphaeriaceae fungi. Identification of 16 Botryosphaeriaceae isolates was carried out based on both morphological characteristics and phylogenetic analyses of combined ITS, LSU, tef1-α, and tub2 sequence data. Four novel species (Dothiorella ovata, Do. rosacearum, Do. septata, and Lasiodiplodia delonicis) and seven previously known species (Botryosphaeria fujianensis, Diplodia mutila, Di. seriata, L. crassispora, L. mahajangana, Macrophomina euphorbiicola and Sphaeropsis eucalypticola) were identified while new hosts and geographical records were reported. This study indicates that the fungal family Botryosphaeriaceae seems to be common and widespread on a broad range of hosts in China and Thailand.

Morpho-Phylogenetic Evidence Reveals Novel Species and New Records of Botryosphaeriaceae in China and Thailand

Celtis sinensis is an adaptable species that is widely grown in southern China. In June 2022, a leaf blotch disease of C. sinensis was observed in Nanjing, Jiangsu, China. Based on morphological characterization, three isolates were identified as Alternaria species. Phylogenetic analysis of combined ITS, GAPDH, TEF1-α, RPB2 and Alt a 1 sequences identified the three isolates we obtained as Alternaria alternata and Alternaria koreana. Koch’s postulates were fulfilled in the greenhouse, and the pathogenicity of A. alternata and A. koreana was determined by leaf inoculation tests on C. sinensis seedlings. The symptoms of indoor inoculation were consistent with those in the field. A. alternata and A. koreana can grow at 15-35 °C, with an optimal growth temperature of 25 °C. The results of fungicide sensitivity experiments indicated that A. alternata and A. koreana were the most sensitive to prochloraz, which may be a useful strategy for the future prevention and control management of A. alternata and A. koreana. This study provides the first step for further research on A. alternata and A. koreana as pathogens of C. sinensis and provides a theoretical basis for future control strategies. 

&lt;em&gt;Alternaria alternata&lt;/em&gt; and A.&lt;em&gt;koreana&lt;/em&gt;, the Causal Agent of a New Leaf Spot in Celtis sinensis and Their Sensitivity to Fungicides

Celtis sinensis is a highly versatile species that is commonly cultivated in the southern regions of China. In June 2022, leaf spot disease was detected in C. sinensis in Nanjing, Jiangsu, China. Based on morphological characteristics, three isolates were determined to be of the Alternaria species. A phylogenetic analysis of combined ITS, GAPDH, TEF1-α, RPB2, and Alt a 1 sequences identified the three isolates we obtained as Alternaria alternata and A. koreana. We determined the pathogenicity of A. alternata and A. koreana in C. sinensis leaves using inoculation through in vivo experiments. Symptoms of inoculation onset in indoor pots were in accordance with those observed in the open country. A. alternata and A. koreana can grow at 15–35 °C, with the best growth occurring at 25 °C. The findings from fungicide susceptibility experiments demonstrated that A. alternata and A. koreana were the most sensitive to prochloraz, which could offer an effective approach for future prevention and control measures against A. alternata and A. koreana. This study provides the initial step toward further research on A. alternata and A. koreana as pathogens of C. sinensis and provides the stage for future control strategy development.

Alternaria alternata and Alternaria koreana, the Causal Agents of Leaf Spot in Celtis sinensis and Their Sensitivity to Fungicides

Abstract Yunnan Province is located in southwestern China, at the core and intersection of the “Himalaya”, “Indo-Burma” and “Mountains of Southwest China” biodiversity hotspots. It is the most biodiverse province in China, acting as a major center for the origin and diversification of numerous species, and of which lignicolous freshwater fungi are one of the rich bioresources. As a part of our ongoing studies on freshwater fungi in Yunnan Province China, we collected lignicolous freshwater ascomycetes from Yunnan plateau lakes, combining morphological characteristics and multi-gene phylogenetic analysis (including, ITS, LSU, SSU, tef 1-α, tub 2 and rpb 2) to identify the species and reveal their phylogenetic placement. A total of 293 freshwater ascomycetes were collected, and through rigorous analysis, 126 species were identified, spanning three classes, 24 orders, 39 families, and 70 genera. Most of these fungi belong to Dothideomycetes and Sordariomycetes , with a few of Eurotiomycetes . Based on morphological and phylogenetic analyses, we introduce two new genera, Neomoromyces and Rostraeuseptisporum , and 40 new species, viz . , Apiospora fuxianhuensis , A. lacustris , Atractospora hydei , Chaetopsina hydei , C. septata , Chloridium hydei , Ch. yunnanense , Dematiosporium hydei , D. muriforme , Dictyocheirospora yunnanensis , Distoseptispora dujuanhuensis , Di. hongheensis , Di. jingdongensis , Halobyssothecium hydei , Hongkongmyces hydei , Kirschsteiniothelia hydei , Mytilinidion hydei , Neomoromyces hydei , Obliquifusoideum hydei , Ophioceras yunnanense , Plagiascoma hydei , Pseudodactylaria lacustris , Pseudostanjehughesia hydei , Rostraeuseptisporum hydei , Sporidesmiella dujuanhuensis , S. guttulata , S. hongheensis , S. hydei , S. lacustris , Sporidesmium dianchiense , Sp. distoseptatum , Sp. dujuanhuense , Sp. hongheense , Sp. lacustris , Sp. kunmingense , Sp. yangzonghaiense , Sp. yilonghuense , Thysanorea hydei , Tetraploa verrucosa , Xylolentia hydei . Detailed morphological descriptions and illustrations of these species are provided, along with a discussion of their phylogenetic relationships and distinctive morphological characters. Furthermore, five new combinations are introduced, viz . , Pseudodactylaria flammulicornuta (≡ Dodactylaria flammulicornuta ), P. palmae (≡ Do. palmae ), P. tunicata (≡ Do. tunicata ), P. uliginicola (≡ Do. uliginicola ) and Pseudostanjehughesia verrucosa (≡ Ceratosporium verrucosum ). Based on molecular sequence data and morphological characteristics, Dictyocheirospora aquadulcis and Dic. lithocarpi have been synonymized with Dic. heptaspora , Dic. alangii synonymized with Dic. appendiculata ; Dictyosporium lakefuxianense synonymized with Pseudodictyosporium wauense , Distoseptispora nanchangensis synonymized with Distoseptispora aquatica , Chaetopsina beijingensis synonymized with Ch. fulva . Ten new geographical records are reported in China and 10 species are first reported from freshwater habitats, and 6 species are newly reported both from China and from freshwater habitats. This study fills a gap in the research on fungal diversity in Yunnan Province, and improves our understanding of their ecological roles in freshwater ecosystems. Phylogenetic analysis provides a reliable molecular framework for the classification of lignicolous freshwater fungi, supporting the reassessment of fungal taxonomy and ensuring a more objective and evolutionarily natural classification of species. This work is dedicated to Professor Kevin D. Hyde on his 70th birthday, in recognition of his lifetime contributions to mycology and his extensive research and training of students. His wide-ranging work on freshwater fungi is highlighted in this paper. 

Taxonomy and phylogeny of lignicolous freshwater fungi from plateau lakes in Yunnan Province, China

Cryptomeria japonica D. Don is a coniferous tree species widely grown in southern China for its high ornamental value. Recently, during disease surveys in China, a symptom of dieback occurred on C. japonica in Nanjing, Jiangsu Province, China. A total of 130 trees were surveyed and more than 90% showed the same symptom. The crowns of affected trees were brown when viewing from a distance, and the bark showed no difference from the healthy ones. In this study, 157 isolates were isolated from the 3 affected plants of C. japonica, and based on the living culture on PDA, the fungal isolates were preliminarily divided into 6 groups. Thirteen representative isolates were selected for the pathogenicity test, and seven of them showed obvious pathogenicity on C. japonica, causing stem basal canker. These isolates were identified based on comparisons of the DNA sequences of the internal transcribed spacer regions (ITS), partial translation elongation factor 1-alpha (tef1), β-tubulin (tub2), and DNA-directed RNA polymerase II subunit (rpb2) and combined with their morphological characteristics. Results showed that these seven isolates belong to two taxa in Neofusicoccum, including a species new to science. The new species, Neofusicoccum cryptomeriae, was hereby described and illustrated. The other species was N. parvum. Both species were pathogens of stem basal canker of Cryptomeria japonica.
