Wave front shaping, the ability to manipulate light fields both spatially and temporally, in complex media is an emerging field with many applications. This review summarizes how insights from mesoscopic scattering theory have direct relevance for optical wave control experiments and vice versa. The results are expected to have an impact on a number of fields ranging from biomedical imaging to nanophotonics, quantum information, and communication technology.

Light fields in complex media: Mesoscopic scattering meets wave control

In the field of biomedical optics, optical scattering has traditionally limited the range of imaging within tissue to a depth of one millimetre. A recently developed class of wavefront-shaping techniques now aims to overcome this limit and achieve diffraction-limited control of light beyond one centimetre. By manipulating the spatial profile of an optical field before it enters a scattering medium, it is possible to create a micrometre-scale focal spot deep within tissue. To successfully operate in vivo, these wavefront-shaping techniques typically require feedback from within the biological sample. This Review summarizes recently developed 'guidestar' mechanisms that provide feedback for intra-tissue focusing. Potential applications of guidestar-assisted focusing include optogenetic control over neurons, targeted photodynamic therapy and deep tissue imaging.

Recent progress in wavefront shaping has enabled control of light propagation inside linear media to focus and image through scattering objects. In particular, light propagation in multimode fibres comprises complex intermodal interactions and rich spatiotemporal dynamics. Control of physical phenomena in multimode fibres and its applications are in their infancy, opening opportunities to take advantage of complex nonlinear modal dynamics. Here, we demonstrate a wavefront shaping approach for controlling nonlinear phenomena in multimode fibres. Using a spatial light modulator at the fibre input, real-time spectral feedback and a genetic algorithm optimization, we control a highly nonlinear multimode stimulated Raman scattering cascade and its interplay with four-wave mixing via a flexible implicit control on the superposition of modes coupled into the fibre. We show versatile spectrum manipulations including shifts, suppression, and enhancement of Stokes and anti-Stokes peaks. These demonstrations illustrate the power of wavefront shaping to control and optimize nonlinear wave propagation. The combination of a spatial light modulator at the fibre input, real-time spectral feedback and a genetic algorithm optimization controls the nonlinear stimulated Raman scattering cascade and its interplay with four-wave mixing in multimode fibres.

Adaptive wavefront shaping for controlling nonlinear multimode interactions in optical fibres

A long-standing goal in biomedical imaging, the control of light inside turbid media, requires knowledge of how the phase and amplitude of an illuminating wavefront are transformed as the electric field propagates inside a scattering sample onto a target plane. So far, it has proved challenging to non-invasively characterize the scattered optical wavefront inside a disordered medium. Here, we present a non-invasive scattering compensation method, termed F-SHARP, which allows us to measure the scattered electric-field point spread function (E-field PSF) in three dimensions. Knowledge of the phase and amplitude of the E-field PSF makes it possible to optically cancel sample turbulence. We demonstrate the imaging capabilities of this technique on a variety of samples and notably through vertebrate brains and across thinned skull in vivo. A non-invasive scattering compensation method, termed F-SHARP, gives direct access to the phase and amplitude of the electric-field point spread function, enabling fast and high-resolution correction of aberrations and scattering in living tissue.

Diffraction-limited imaging through complex scattering media is a long-sought-after goal with important applications in biomedical research. In recent years, high-resolution wavefront shaping has emerged as a powerful approach to generate a sharp focus through highly scattering, visually opaque samples. However, it requires a localized feedback signal from the target point of interest, which necessitates an invasive procedure in all-optical techniques. Here, we show that by exploiting optical nonlinearities, a diffraction-limited focus can be formed inside or through a complex sample, even when the feedback signal is not localized. We prove our approach theoretically and numerically, and experimentally demonstrate it with a two-photon fluorescence signal through highly scattering biological samples. We use the formed focus to perform two-photon microscopy through highly scattering, visually opaque layers.
