Structures of a diverse set of colchicine binding site inhibitors in complex with tubulin provide a rationale for drug discovery.
Yuxi Wang,Hang Zhang,Benoît Gigant,Yamei Yu,Yangping Wu,Xiangzheng Chen,Qinhuai Lai,Zhaoya Yang,Qiang Chen,Jinliang Yang +9 more
TL;DR: These studies provide a solid structural basis for developing new anti‐cancer agents for the colchicine binding site and show that the binding modes of the CBSIs were different from previous docking models, highlighting the importance of crystal structure information in structure‐based drug design.

Malignant ascites-derived exosomes promote proliferation and induce carcinoma-associated fibroblasts transition in peritoneal mesothelial cells.
Mingtian Wei,Tinghan Yang,Xiangzheng Chen,Yangping Wu,Xiangbing Deng,Wanbin He,Jinliang Yang,Ziqiang Wang +7 more
TL;DR: Peritoneal MCs showed increased ability to proliferate and migrate under the treatment of ascites-derived exosomes, and expression of CAFs specific proteins markers in MCs, including fibroblast activation protein (FAP), alpha-smooth muscle actin (α-SMA), and fibronectin, were increased after treatment of exosome.

Mechanism of microtubule stabilization by taccalonolide AJ.
Yuxi Wang,Yamei Yu,Guo-Bo Li,Shu Ang Li,Chengyong Wu,Benoît Gigant,Wenming Qin,Hao Chen,Yangping Wu,Qiang Chen,Jinliang Yang +10 more
TL;DR: This work proposes that the β-tubulin E-site is locked into a GTP-preferred status by AJ binding, and provides experimental evidence for the connection between MSA binding and tubulin nucleotide state, and will help design new MSAs to overcome taxane resistance.

Isoliquiritigenin prevents the progression of psoriasis-like symptoms by inhibiting NF-κB and proinflammatory cytokines
Yangping Wu,Xiangzheng Chen,Xiaojun Ge,Hongwei Xia,Yuxi Wang,Siyuan Su,Wenting Li,Tinghan Yang,Mingtian Wei,Hang Zhang,Lantu Gou,Jiong Li,Xian Jiang,Jinliang Yang +13 more
TL;DR: ISL exhibited anti-inflammatory effects in psoriasis models, by downregulating IL-6 and IL-8 via suppression of NF-κB activity, suggesting that ISL might serve as a potential candidate for treatment of Psoriasis and other autoimmune inflammatory diseases.

Targeting of DDR1 with antibody-drug conjugates has antitumor effects in a mouse model of colon carcinoma.
Yiran Tao,Ruixue Wang,Qinhuai Lai,Mengdan Wu,Yuxi Wang,Xiaohua Jiang,Lishi Zeng,Shijie Zhou,Zhongping Li,Tinghan Yang,Yuqin Yao,Yangping Wu,Lin Yu,Yuyin Fu,Weirong Lai,Yujia Peng,Ying Lu,Zhixiong Zhang,Cuiyu Guo,Guangbing Zhang,Lantu Gou,Jinliang Yang +21 more
TL;DR: T4H11‐DM4, an ADC targeting DDR1 which carries the tubulin inhibitor payload DM4, highlights the potential of DDR1‐targeted ADC and may facilitate the development of a new effective therapeutic strategy for colon cancer.
