A marker-free Bt transgenic rice line, mfb-MH86, was recently developed in China, which contains a cry1Ab gene driven by a ubiquitin promoter. This Bt gene confers resistance to a range of lepidopteran species, including the striped stem borer, Chilo suppressalis (Walker). The expression of Cry1Ab protein in mfb-MH86 leaves, stems and leaf sheaths (hereinafter referred to as stems), and roots was evaluated throughout the rice-growing season using an enzyme-linked immunosorbent assay. In addition, mfb-MH86 resistance to C. suppressalis, a major pest of rice, was evaluated in a laboratory bioassay with field-collected rice stems. Cry1Ab protein levels of mfb-MH86 were highest in leaves (9.71–34.09 µg/g dry weight [DW]), intermediate in stems (7.66–18.51 µg/g DW), and lowest in roots (1.95–13.40 µg/g DW). Expression of Cry1Ab Protein in a Marker-Free Transgenic Bt Rice Line and Its Efficacy in Controlling a Target Pest, Chilo suppressalis (Lepidoptera: Crambidae)

Tuta absoluta is a devastating invasive pest worldwide, causing severe damage to the global tomato industry. It has been recorded recently in the northwestern border areas of China, posing a significant threat to tomato production. It was presumed that the region's winter-related low temperatures would avert the alien species from successfully overwintering. In this study, the supercooling capacity and low-temperature tolerance of this pest were examined under laboratory conditions and its overwintering potential in Xinjiang was estimated. The results showed that the lowest supercooling point was recorded in the adult stage (− 19.47 °C), while the highest (− 18.11 °C) was recorded in the pupal stage. The supercooling points of pupae and adults were not influenced by gender. The Ltemp50 and Ltemp90 of female and male adults were the lowest when exposed to cold for 2 h. However, when the duration of exposure extended from 4 to 10 h, the Ltemp50 and Ltemp90 of female and male pupae were the lowest. Comparison of the lowest Ltemp50 and Ltemp90 with temperatures in January indicated that T. absoluta might not be able to overwinter in most of the northern and central regions of Xinjiang. However, in the southern regions, the extremely low temperature was higher than the Ltemp90, suggesting that T. absoluta has a higher overwintering potential in these regions. These results form a basis for predicting the dispersal potential and possible geographic range of this pest in Xinjiang. In addition, our findings provide guidance for the control of this pest by reducing overwintering shelters.

Supercooling capacity and cold tolerance of the South American tomato pinworm, Tuta absoluta, a newly invaded pest in China

Although parasitoids ultimately kill their host, koinobiont parasitoids must protect not only themselves but also their hosts against extreme environments. In this study, the parasitism rate of Chilo suppressalis Walker (Lepidoptera: Pyralidae) was investigated, and the average body weights, supercooling points, and concentrations of glycerol (acting as a cryoprotectant) in the hemolymph were compared between parasitized and non-parasitized larvae. Five species of koinobiont endoparasitoids parasitized the overwintering C. suppressalis larvae and the total parasitism rate was 47.6% (n = 1 537). Average body weight of parasitized larvae was significantly lower than that of non-parasitized larvae, and the parasitism rate of the lighter group (20–30 mg) was highest. The supercooling point of parasitized C. suppressalis larvae (−15.7 ± 0.3 °C) was significantly lower than that of the non-parasitized larvae (−14.3 ± 0.2 °C). In addition, supercooling points were not correlated with body weights between parasitized and non-parasitized larvae, indicating that cold hardiness of parasitized larvae was enhanced by endoparasitoids. Furthermore, the concentration of glycerol in the hemolymph was significantly higher in parasitized larvae (205.0 ± 7.1 μmol ml−1) than in non-parasitized larvae (169.8 ± 14.4 μmol ml−1), which suggests that the mechanism that decreases the supercooling point of parasitized larvae was associated with glycerol. All these results indicated that the cold hardiness of parasitized C. suppressalis larvae was enhanced by their endoparasitoids, which benefitted overwintering endoparasitoids.

Overwintering strategy of endoparasitoids in Chilo suppressalis: a perspective from the cold hardiness of a host

The Chinese citrus fruit fly, Bactrocera minax (Enderlein), is an economically important pest of citrus. The fly has an obligatory pupal diapause in soil from November to March. However, techniques for predicting or determining the emergence of the adult have, thus far, not been well documented. In this study, we investigated the effects of different pre-diapause temperatures (8, 12, 16, and 20°C) and pupal body weight (five groups according to pupal weight: G-58, 55.0-61.0 mg; G-68, 65-71 mg; G-78, 75-81 mg; G-88, 85-91 mg; G-95, 92-98 mg) on pupal period (the indicator of diapause intensity). The pupal period of B. minax larvae pupated at 8°C was 193.41 d, which was significantly shorter than that of larvae incubated at higher temperatures, suggesting that there was a lower diapause intensity for larvae pupated at lower pre-diapause temperatures. There were also significant differences in the pupal periods at different pupal body weights. The pupal period of G-58 was significantly shorter than that of the heavier groups (G-88 and G-95), and the pupal period increased with increasing pupal body weight in the five groups. Moreover, the pupal period of B. minax significantly and positively correlated to pupal body weight. These findings demonstrate that the pre-diapause temperature and pupal body weight are suitable indicators for predicting the pupal period of overwintering individuals, and the results of this study will contribute to the development of new and effective strategies for predicting the occurrence and population dynamics of B. minax adult.
