The Biomarkers of Nutrition for Development (BOND) project is designed to provide evidence-based advice to anyone with an interest in the role of nutrition in health. Specifically, the BOND program provides state-of-the-art information and service with regard to selection, use, and interpretation of biomarkers of nutrient exposure, status, function, and effect. To accomplish this objective, expert panels are recruited to evaluate the literature and to draft comprehensive reports on the current state of the art with regard to specific nutrient biology and available biomarkers for assessing nutrients in body tissues at the individual and population level. Phase I of the BOND project includes the evaluation of biomarkers for 6 nutrients: iodine, iron, zinc, folate, vitamin A, and vitamin B-12. This review represents the second in the series of reviews and covers all relevant aspects of folate biology and biomarkers. The article is organized to provide the reader with a full appreciation of folate's history as a public health issue, its biology, and an overview of available biomarkers (serum folate, RBC folate, and plasma homocysteine concentrations) and their interpretation across a range of clinical and population-based uses. The article also includes a list of priority research needs for advancing the area of folate biomarkers related to nutritional health status and development.

Genome-wide DNA methylation analysis identifies hypomethylated genes regulated by FOXP3 in human regulatory T cells

Nonalcoholic fatty liver disease (NAFLD) refers to a wide spectrum of liver disease that is not from excess alcohol consumption, but is often associated with obesity, type 2 diabetes, and metabolic syndrome. NAFLD pathogenesis is complicated and involves oxidative stress, lipotoxicity, mitochondrial damage, insulin resistance, inflammation, and excessive dietary fat intake, which increase hepatic lipid influx and de novo lipogenesis and impair insulin signaling, thus promoting hepatic triglyceride accumulation and ultimately NAFLD. Overproduction of proinflammatory adipokines from adipose tissue also affects hepatic metabolic function. Current NAFLD therapies are limited; thus, much attention has been focused on identification of potential dietary substances from fruits, vegetables, and edible plants to provide a new strategy for NAFLD treatment. Dietary natural compounds, such as carotenoids, omega-3-PUFAs, flavonoids, isothiocyanates, terpenoids, curcumin, and resveratrol, act through a variety of mechanisms to prevent and improve NAFLD. Here, we summarize and briefly discuss the currently known targets and signaling pathways as well as the role of dietary natural compounds that interfere with NAFLD pathogenesis.

Chemoprevention of nonalcoholic fatty liver disease by dietary natural compounds.

Systemic lupus erythematosus (SLE) is a complicated autoimmune disease of multifactorial pathoaetiology. One of the most serious manifestations is lupus nephritis. The pathogenesis of SLE has not been well elucidated, but it has been reported that interleukin-17 (IL-17) and Th17 cells play important roles in the pathogenesis of SLE. IL-17A, a member of IL-17 family, amplifies the immune response by inducing the local production of chemokines and cytokines, recruiting neutrophils and monocytes, augmenting the production of autoantibodies, and aggravating the inflammation and damage of target organs such as the kidney in SLE. In recent years, several IL-17A pathway inhibitors have advanced into clinical trials, including the anti-IL-17A monoclonal antibody and the anti-17RA monoclonal antibody. Interleukin-17 in systemic lupus erythematosus: A comprehensive review

Docosahexaenoic acid (DHA) is one kind of ω-3 polyunsaturated fatty acids (PUFAs) and plays an important role in lipid metabolism. In this research, mice were daily intragastric administrated with DHA for 3 weeks. Subcutaneous adipose tissue and liver were separated every week, RNA was extracted. Peroxisome proliferator-activated receptor (PPARγ), Sterol regulatory element binding protein-1c (SREBP-1c), Fatty acid synthetase (FAS), Hormone sensitive lipase (HSL) and triglyceride hydrolase TGH genes expression were detected by quantitative PCR. Data showed that, DHA up-regulated PPARγ, HSL and TGH in adipose tissue, but it had no effect on SREBP-1c and FAS expression. However, in liver there were some differences in regulating these genes. PPARγ, SREBP-1c and FAS were down-regulated, HSL was up-regulated and TGH had no change. These results indicated that DHA played different regulating roles in lipid metabolism in different tissues. In adipose tissue, DHA increased the expression of lipogenesis and lipolysis genes. In liver lipogenesis genes were decreased, but lipolysis genes were increased by DHA. In conclusion, DHA could reduce body fat mass through regulating lipogenesis and lipolysis genes.

Circulating CD4+ T helper cells are activated through interactions with antigen presenting cells and undergo differentiation into specific T helper cell subsets depending on the type of antigen encountered. In addition, the relative composition of the circulating CD4+ T cell population changes as animals mature with an increased percentage of the population being memory/effector type cells.

Aging and dysregulation of immune responds are closely associated through a complicated but unclear mechanism. Although many theories have been proposed as overall dysregulation involved in aging, mechanisms such as efficiency of DNA repairing, over-expression of transcription factors (such as NF-κB family), and shift of cell types, are among many factors that contribute to and affect aging process. It is of great interests to understand the possible mechanism that is involved in aging immune system. Here, we report that the inducible genes Il2 and Csf2 are increased as T cells undergo activation and aging. Of particular note were the findings that the relative composition of the circulating CD4(+) T cell population changes as animals mature with an increased percentage of the population being memory/effector type cells. In addition, mRNA levels of NF-κB family genes that are essential elements for cytokine activation in adult mice and activated T cells are significantly increased. We have demonstrated that the expression of inducible genes is accompanied by increased memory/effector type cells and by increased expression level of NF-κB family genes during cell activation and development.
