The lion's share of bacteria in various environments cannot be cloned in the laboratory and thus cannot be sequenced using existing technologies. A major goal of single-cell genomics is to complement gene-centric metagenomic data with whole-genome assemblies of uncultivated organisms. Assembly of single-cell data is challenging because of highly non-uniform read coverage as well as elevated levels of sequencing errors and chimeric reads. We describe SPAdes, a new assembler for both single-cell and standard (multicell) assembly, and demonstrate that it improves on the recently released E+V-SC assembler (specialized for single-cell data) and on popular assemblers Velvet and SoapDeNovo (for multicell data). SPAdes generates single-cell assemblies, providing information about genomes of uncultivatable bacteria that vastly exceeds what may be obtained via traditional metagenomics studies. SPAdes is available online ( http://bioinf.spbau.ru/spades ). It is distributed as open source software.

To understand the effects of temperature on biological systems, we compile, organize, and analyze a database of 1,072 thermal responses for microbes, plants, and animals. The unprecedented diversity of traits (n = 112), species (n = 309), body sizes (15 orders of magnitude), and habitats (all major biomes) in our database allows us to quantify novel features of the temperature response of biological traits. In particular, analysis of the rising component of within-species (intraspecific) responses reveals that 87% are fit well by the Boltzmann–Arrhenius model. The mean activation energy for these rises is 0.66 ± 0.05 eV, similar to the reported across-species (interspecific) value of 0.65 eV. However, systematic variation in the distribution of rise activation energies is evident, including previously unrecognized right skewness around a median of 0.55 eV. This skewness exists across levels of organization, taxa, trophic groups, and habitats, and it is partially explained by prey having increased trait performance at lower temperatures relative to predators, suggesting a thermal version of the life-dinner principle—stronger selection on running for your life than running for your dinner. For unimodal responses, habitat (marine, freshwater, and terrestrial) largely explains the mean temperature at which trait values are optimal but not variation around the mean. The distribution of activation energies for trait falls has a mean of 1.15 ± 0.39 eV (significantly higher than rises) and is also right-skewed. Our results highlight generalities and deviations in the thermal response of biological traits and help to provide a basis to predict better how biological systems, from cells to communities, respond to temperature change.

Diamondback Moth Ecology and Management: Problems, Progress, and Prospects

A new classification of lifestyles of phytoseiid mites is proposed based on the findings and suggestions from many studies conducted in the fifteen years since the publication of the lifestyle system by McMurtry & Croft (1997). In this newly proposed classification, Type I (Specialized mite predators) is divided into three subtypes to highlight mite prey specificity: subtype I-a—Specialized predators of Tetranychus species (Tetranychidae); subtype I-b—Specialized predators of web-nest producing mites (Tetranychidae); subtype I-c—Specialized predators of tydeoids (Tydeoidea). Type II (Selective predators of tetranychid mites) remains essentially the same. Type III (Generalist predators) is a huge and diverse group of general feeders; predators in this category are now grouped into five subtypes based on the microhabitat occupied and morphology: subtype III-a— Generalist predators living on pubescent leaves; subtype III-b—Generalist predators living on glabrous leaves; III-c—Generalist predators living in confined spaces on dicotyledonous plants; III-d—Generalist predators living in confined spaces on monocotyledonous plants; III-e—Generalist predators from soil/litter habitats. Type IV (Pollen feeding generalist predators) also remains essentially the same. Two possible additional lifestyles include phytoseiids living on aquatic plants and phytoseiids able to pierce leaf cells. Behavioral and biological information is updated and implications for biological control strategies are discussed.

The chemokine ligands probably comprise one of the first completely known molecular superfamilies. The genomic organization of the chemokine ligand genes and a comparison of their sequences between species shows that tandem gene duplication has taken place independently in the mouse and human lineages of some chemokine families. This means that care needs to be taken when extrapolating experimental results on some chemokines from mouse to human.

Abstract The effectiveness of predators in suppressing prey populations is known to be influenced by the predator-prey release ratio. The efficacy of Neoseiulus cucumeris (Oudemans) against Frankliniella occidentalis (Pergand) in the presence or absence of Tetranychus urticae Koch and Phytoseiulus persimilis Athias-Henriot on rose plants was evaluated in the present study. The decrease of F. occidentalis populations was compared at the six predator:prey ratios of 1:2, 1:5, 1:10, 1:20, 1:50, and 1:100 in the laboratory, and at the three release rates of 20, 40, and 100 predators per m2 in microcosm conditions. The plants without predators were considered as controls. All treatments with the release of N. cucumeris resulted in a significant decrease in F. occidentalis larval population compared to the control. The most suppression (84.5%) of F. occidentalis larvae was observed in the release rate of 100 adults of N. cucumeris per m2 in 30 days after the release of the predator. Predator ratios of 1:2 and 1:5 of N. cucumeris: F. occidentalis resulted in a 94.6 and 93.4% decrease in F. occidentalis larvae after 24 hours, respectively. The presence of T. urticae (as a competitor herbivore) and P. persimilis had no effect on the efficiency of N. cucumeris in the suppression of F. occidentalis. The dense webs of T. urticae decreased the efficiency of N. cucumeris at predator:prey ratios of 1:5 and 1:10 more than1:2. This study showed that the release of 100 adults of N. cucumeris per m2 would be sufficient to control F. occidentalis larvae on rose plants. Moreover, we demonstrated that higher release rates of N.cucumeris are needed when T. urticae's dense webs are present.
