VCAM-1 promotes osteolytic expansion of indolent bone micrometastasis of breast cancer by engaging α4β1-positive osteoclast progenitors
Xin Lu,Euphemia Mu,Yong Wei,Sabine Riethdorf,Qifeng Yang,Qifeng Yang,Min Yuan,Jun Yan,Yuling Hua,Benjamin Tiede,Xuemin Lu,Bruce G. Haffty,Klaus Pantel,Joan Massagué,Yibin Kang,Yibin Kang +15 more
TL;DR: A bone metastasis dormancy model is characterized to show that aberrant expression of vascular cell adhesion molecule 1 (VCAM-1), in part dependent on the activity of the NF-κB pathway, promotes the transition from indolent micrometastasis to overt metastasis.

Myeloid-derived suppressor cells inhibit T cell activation through nitrating LCK in mouse cancers
Shan Feng,Shan Feng,Xi Cheng,Xi Cheng,Lin Zhang,Xuemin Lu,Seema Chaudhary,Ruifang Teng,Christian Frederickson,Matthew M. Champion,Ren Zhao,Liang Cheng,Yiyi Gong,Haiteng Deng,Xin Lu,Xin Lu +15 more
TL;DR: In this paper, the authors applied a nitroproteomic approach based on chemical derivation of 3-nitrotyrosine and identified that lymphocyte-specific protein tyrosine kinase (LCK) is nitrated at Tyr394 by myeloid-derived suppressor cells (MDSCs).

Neutrophils resist ferroptosis and promote breast cancer metastasis through aconitate decarboxylase 1.
Yun Zhao,Zhongshun Liu,Guoqiang Liu,Yuting Zhang,Sheng Liu,Dailin Gan,Wennan Chang,Xiaoxia Peng,Eun Suh Sung,Keegan Gilbert,Yini Zhu,Xuechun Wang,Ziyu Zeng,Hope Baldwin,Guanzhu Ren,Jessica Weaver,Anna Huron,Toni Mayberry,Qingfei Wang,Yujue Wang,M. Elena Díaz-Rubio,Xiaoyang Su,M. S. Stack,Siyuan Zhang,Xuemin Lu,R. Sheldon,Jun Li,Chi Zhang,Jun Wan,Xin Lu +29 more
TL;DR: These findings reveal how TINs escape from ferroptosis through the Acod1-dependent immunometabolism switch and establish Acod 1 as a target to offset immunosuppression and improve immunotherapy against metastasis.

TL;DR: A nitroproteomic approach is developed and a potential mechanism for the immunotherapy resistance where a key protein for T cell activation, lymphocyte-specific protein tyrosine kinase (LCK), is nitrated and inactivated by myeloid-derived suppressor cell–generated reactive nitrogen species (RNS) in the resistant tumor is uncovered.

Opposing roles of TGFβ and BMP signaling in prostate cancer development.
Xin Lu,Xin Lu,Xin Lu,Eun-Jung Jin,Eun-Jung Jin,Xi Cheng,Xi Cheng,Shan Feng,Shan Feng,Xiaoying Shang,Pingna Deng,Shan Jiang,Qing Chang,Sharif Rahmy,Seema Chaudhary,Xuemin Lu,Ren Zhao,Y. Alan Wang,Ronald A. DePinho +18 more
TL;DR: It is demonstrated that the molecular actions of TGFBR2 result in both SMAD4-dependent constraint of proliferation and SMAD1-independent activation of apoptosis, and BMPR2 deletion extended survival relative to Pten deletion alone, establishing its promoting role in BMP6-driven prostate cancer progression.
