Plants produce a variety of secondary metabolites in response to biotic and abiotic stresses. Although they have many functions, a subclass of toxic secondary metabolites mainly serve plants as deterring agents against herbivores, insects, or pathogens. Microorganisms present in divergent ecological niches, such as soil, water, or insect and rumen gut systems have been found capable of detoxifying these metabolites. As a result of detoxification, microbes gain growth nutrients and benefit their herbivory host via detoxifying symbiosis. Here, we review current knowledge on microbial degradation of toxic alkaloids, glucosinolates, terpenes, and polyphenols with an emphasis on the genes and enzymes involved in breakdown pathways. We highlight that the insect-associated microbes might find application in biotechnology and become targets for an alternative microbial pest control strategy.

Microbial degradation of plant toxins.

Soil aggregation contributes to the stability of soil structure and the sequestration of soil organic carbon (SOC), making it an important indicator of soil health in agroecosystems. Crop diversification is considered a rational management practice for promoting sustainable agriculture. However, the complexity of cropping systems and crop species across different regions limits our comprehensive understanding of soil aggregation and associated carbon (C) content under crop diversification. Therefore, we conducted a meta-analysis by integrating 1924 observations from three diversification strategies (cover crops, crop rotation, and intercropping) in global agroecosystems to explore the effects of crop diversification on soil aggregates and associated C content. The results showed that compared to monoculture, crop diversification significantly increased the mean weight diameter and bulk soil C by 7.5% and 3.3%, respectively. Furthermore, there was a significant increase in the proportion of macroaggregates and their associated C content by 5.0% and 12.5%, while there was a significant decrease in the proportion of microaggregates as well as silt-clay fractions along with their associated C under crop diversification. Through further analysis, we identified several important factors that influence changes in soil aggregation and C content induced by crop diversification including climatic conditions, soil properties, crop species, and agronomic practices at the experimental sites. Interestingly, no significant differences were found among the three cropping systems (cover crops, crop rotation, and intercropping), while the effects induced by crop diversifications showed relatively consistent results for monoculture crops as well as additive crops and crop diversity. Moreover, the impact of crop diversification on soil aggregates and associated C content is influenced by soil properties such as pH and SOC. In general, our findings demonstrate that crop diversification promotes soil aggregation and enhances SOC levels in agroecosystems worldwide. 

Crop diversification promotes soil aggregation and carbon accumulation in global agroecosystems: A meta-analysis.

Plant autotoxicity is considered to be one of the important causes of continuous cropping obstacles in modern agriculture, which accumulates a lot of allelochemicals and xenobiotics and is difficult to solve effectively. To overcome tobacco continuous obstacles, a strain Pigmentiphaga kullae CHJ604 isolated from the environment can effectively degrade these compounds in this study. CHJ604 strain can degrade 11 types of autotoxicity allelochemicals and xenobiotics (1646.22 μg/kg) accumulated in the soil of ten-years continuous cropping of tobacco. The 11 allelochemicals and xenobiotics significantly reduced Germination Percentage (GP), Germination Index (GI), and Mean Germination Time (MGT) of tobacco seeds, and inhibited the development of leaves, stems, and roots. These negative disturbances can be eliminated by CHJ604 strain. The degradation pathways of 11 allelochemicals and xenobiotics were obtained by whole genome sequence and annotation of CHJ604 strain. The heterologous expression of a terephthalate 1,2-dioxygenase can catalyze 4-hydroxybenzoic acid, 4-hydroxy-3-methoxybenzoic acid, 4-hydroxybenzaldehyde, and 4-hydroxy-3-methoxy-benzaldehyde, respectively. The phthalate 4,5-dioxygenase can catalyze phthalic acid, diisobutyl phthalate, and dibutyl phthalate. These two enzymes are conducive to the simultaneous degradation of multiple allelochemicals and xenobiotics by strain CHJ604. This study provides new insights into the biodegradation of autotoxicity allelochemicals and xenobiotics as it is the first to describe a degrading bacterium of 11 types of allelochemicals and xenobiotics and their great potential in improving tobacco continuous obstacles.

Long-term continuous cropping of tobacco causes dysbiosis of soil microbial communities, the imbalance of soil nutrients, and the increase of pathogenic bacteria, which will slow the growth and development of tobacco plants, reduce the production quality, and cause significant losses to tobacco production and tobacco farmers. The application of Potassium fulvic acid can not only provide nutrients, but also inhibit the propagation of pathogens in soil along with raising the amount of organic matter in the soil, which is an effective way to improve soil health. In this experiment, Tobacco variety SNT60 was used as the test material, and 6 treatments were set up by pot test, they were: no fertilisation control group (CK), tobacco special fertiliser (NPK), 3.45 g/kg of potassium fulvic acid fertiliser (T1), 4.65 g/kg of potassium fulvic acid fertiliser (T2), 5.85 g/kg of potassium fulvic acid fertiliser (T3), 7.05 g/kg of potassium fulvic acid fertiliser (T4), Ten replications were set up for each treatment and the soil and fertiliser were mixed and potted before transplanting, 70% as basal fertiliser and 30% as supplementary fertiliser. We also analyzed soil properties, soil microorganisms and agronomic traits of tobacco plants in different treatments to provide reference for mitigating tobacco succession barrier. The test results are as follows: 4.65 g/kg of potassium fulvic acid fertiliser (T2) treatment was the best, soil organic matter, quick nitrogen, phosphorus, potassium, pH, soil catalase, soil sucrase, and soil urease content, compared to CK control, increased by 22.04%, 43.12%, 96.21%, 381.79%, 25.43%, 91.69%, 262.07% and 93.16%. In terms of microbial community, application of potassium fulvic acid fertiliser significantly increased the relative abundance of Ascomycetes, Chlorobacterium, Bacillus, Proteobacteria and Tephritobacterium in the soil. Meanwhile, 4.65 g/kg of potassium fulvic acid fertiliser (T2) promoted the growth of tobacco plants, improved leaf photosynthetic capacity, and enhanced plant disease resistance. This experiment provides practical measures to improve the microbial community of tobacco continuous cropping soils and to reduce the incidence of diseases. 

Effect of potassium fulvate on continuous tobacco cropping soils and crop growth

. Tobacco Bacterial Wilt (TBW) is a disease caused by the bacterium Ralstonia solanacearum . R. solanacearum is a soil-borne plant pathogen that can cause devastating losses in agricultural crops. However, there is limited information on the variation in the microbial community structure and molecular characteristics in soils with different index levels of tobacco bacterial wilt. Therefore, current study focused at undertaking molecular analysis of microbial community characteristics in soils of different index levels of TBW with the aim of providing insights on bio-control opportunities for bacterial wilt incidence in agricultural farms for improved yield. The findings indicated that the genus Enterobacteriaceae from the phylum Proteobacteria was more abundant under the control group (healthy soils), on the other hand genus Ralstonia of the phylum Proteobacteria was more abundant in the JYRS9 group (highly infected soils). The use of sample similarity analysis and COG function classification in relation to tobacco wilt disease index provided a comprehensive understanding of the microbial ecology of soil and the conditions that influence disease development and management. the use of KEGG pathway analysis in relation to tobacco wilt disease index provided a deeper understanding of the microbial ecology of soil and the factors that influence disease development and management.

Molecular analysis of microbial community characteristics in soils with different levels of tobacco bacterial wilt indices

Abstract Long-term continuous monoculture cropping of tobacco leads to high incidence of tobacco bacterial wilt (TBW) caused by Ralstonia solanacearum , which threatening world tobacco production and causing great economy loss. In this study, a safe and effective way to control TBW by microbial degradation of phenolic allelochemicals (PAs) was explored. Eleven kinds of PAs were identified from continuous tobacco cropping soil. These PAs exhibited various effects on the growth, chemotaxis and biofilm formation of R . solanacearum . Then we isolated eight strains of Bacillus , one strain of Brucella, one strain of Enterobacter and one strain of Stenotrophomonas capable of degrading these PAs. The results of degradation assay showed that these isolated strains could degrade PAs both in culture solutions and soil. Besides, the incidence of TBW caused by R . solanacearum and deteriorated by PAs were significantly decreased by treating with these degrading strains. Furthermore, six out of eleven isolated strains were combined to degrade all the identified PAs and ultimately sharply reduced the incidence of TBW by 61.44% in pot experiment. In addition, the combined degrading bacteria could promote the plant growth and defense response. This study will provide a promising strategy for TBW control in tobacco production.
