Spatial heterogeneity of infiltrating T cells in high-grade serous ovarian cancer revealed by multi-omics analysis
Bin Yang,Xiong Li,Wei Zhang,Junpeng Fan,Yong-guo Zhou,Wenting Li,Jin Gou Yin,Xiaohang Yang,Ensong Guo,Xi Li,Siying Liu,Dianxing Hu,Xu Qin,Yingyu Dou,Rourou Xiao,Funian Lu,Zizhuo Wang,Tianyu Qin,Wei Wang,Qinghua Zhang,Shuai Cheng Li,Ding Ma,Gordon B. Mills,Gang Chen,Chaoyang Sun +24 more
TL;DR: In this paper , the authors integrated RNA sequencing, whole-genome sequencing, bulk T cell receptor (TCR) sequencing, as well as single-cell RNA/TCR sequencing to investigate the characteristics and differential composition of TILs across different HGSOC sites.

WEE1 inhibition induces anti-tumor immunity by activating ERV and the dsRNA pathway.
Ensong Guo,Rourou Xiao,Yifan Wu,Funian Lu,Chen Liu,Bin Yang,Xi Li,Yu Fu,Zizhuo Wang,Yuan Li,Yuhan Huang,Fuxia Li,Xue Wu,Lixin You,Tianyu Qin,Yiling Lu,Xiaoyuan Huang,Ding Ma,Gordon B. Mills,Chaoyang Sun,Gang Chen +20 more
TL;DR: The authors showed that targeting WEE1 up-regulates immune signaling through the double-stranded RNA (dsRNA) viral defense pathway with subsequent responsiveness to immune checkpoint blockade even in cGAS/STING-deficient tumors, which is a typical phenotype across multiple cancer types.

Systems approach to rational combination therapy: PARP inhibitors.
TL;DR: A series of studies strongly support the efficacy of the combinatorial adaptive response therapy (CART) platform, wherein targeting the emerging adaptive responses to PARPi with MEK/ERK inhibitors, WEE1/ATR inhibition, and PI3K/AKT/mTOR inhibition showed promising anti-tumor activity in various preclinical models.
BRD4 Inhibition by AZD5153 Promotes Antitumor Immunity via Depolarizing M2 Macrophages.
Xi Li,Yu Fu,Bin Yang,Ensong Guo,Yifan Wu,Jia Huang,Xiaoxiao Zhang,Rourou Xiao,Kezhen Li,Beibei Wang,Junbo Hu,Chaoyang Sun,Gang Chen +12 more
TL;DR: It is clarified that besides eliminating tumor cells directly, AZD5153 works as a regulator of the TAM phenotype, providing a novel strategy combining AZD 5153 and PD-1/PD-L1 antibody that could benefit HGSOC patients.
Inhibiting the IRE1α Axis of the Unfolded Protein Response Enhances the Antitumor Effect of AZD1775 in TP53 Mutant Ovarian Cancer
Rourou Xiao,Lixin You,Li Zhang,Ensong Guo,Faming Zhao,Bin Yang,Xi Li,Funian Lu,Zizhuo Wang,Chen Liu,Wen-Ju Peng,Wenting Li,Xiaohang Yang,Yingyu Dou,Jing-Bing Liu,Wei Wang,Tianyu Qin,Xiao-Xiao Zhang,Fuxia Li,Yang Jin,Qingping Zeng,Beibei Wang,Gordon B. Mills,Gang Chen,Xia Sheng,Chaoyang Sun +25 more
TL;DR: The data reveal an important dual role of the UPR signaling network in mtTP53 ovarian cancer models in response to AZD 1775 and suggest that inhibition of the IRE1α‐XBP1s pathway may enhance the efficacy of AZD1775 in the clinics.
