Motor patterns of the canine gastroduodenal junction were investigated by electromyography under different secretory conditions: enhancement by pentagastrin and reduction by proglumide, cimetidine or CMN 131. The intravenous administration of these antisecretory agents did not affect the spiking activity or the slow-wave frequency of the antrum and duodenum, with the exception of CMN 131 which induced duodenal hyperactivity.

A thiocarboxamide derivative, pyridyl-2-thioacetamide, referred to as CMN 131, was characterized for its gastric antisecretory properties in rats and dogs. Its activity was first determined in 4-hr pylorus-ligated rats and in gastric fistula rats under basal and carbachol-stimulated (18 μg/kg/hr s.c.) conditions. ED50 of acid output inhibition was 1.46 (0.92–2.31) mg/kg by intraduodenal route and 3.85 (2.65–5.59) mg/kg s.c. respectively. The activity was further confirmed in Heidenhain-pouch dogs whose gastric secretion was stimulated by pentagastrin (3.20 μg/kg/hr i.v.) and histamine (160 μg/kg/hr i.v.). Under all these conditions, CMN 131 in doses ranging from 1 to 10 mg/kg, whatever the drug administration route, possesses a long-acting antisecretory property characterized first by an activity on acid concentration and second by an equal inhibitory potency on histamine and pentagastrin stimulation.

The gastric antisecretory activity of pyridyl-2-thioacetamide (CMN 131)

Pyridyl-2-thioacetamide-CMN 131-possess high ulcer-protective activity which seems closely related to its gastric antisecretory properties previously described. This activity is demonstrated using experimental models whose ulcerogenic stimulus can be related to various origins: 4-hr and 24-hr restraint stress ulcers and aspirin or phenylbutazone gastric erosions in rats, histamine-induced ulcers in guinea pigs, reserpine-induced ulcers in cats. In the same dose range used in the ulcer study, and even at higher doses, CMN 131 fails to show any systemic activity on the cholinergic or adrenergic pathway. At a systemic level no antihistaminic, antiserotonin, or ganglion properties can be observed either. Furthermore, CMN 131 modifies neither the salivary and lacrymal secretions, nor the gastric emptying or the blood pressure. Finally, CMN 131 can be considered an original pharmacological agent whose wide antiulcer and antisecretory properties could enable us to understand more clearly the intrinsic mechanism of gastric secretion.
