Polyadenylation of Histone H3.1 mRNA Promotes Cell Transformation by Displacing H3.3 from Gene Regulatory Elements.
Danqi Chen,Qiao Yi Chen,Zhenjia Wang,Yusha Zhu,Thomas Kluz,Wuwei Tan,Jinquan Li,Feng Wu,Lei Fang,Xiaoru Zhang,Rongquan He,Steven S. Shen,Hong Sun,Chongzhi Zang,Chunyuan Jin,Max Costa +15 more
TL;DR: Replication-dependent canonical histone messenger RNAs (mRNAs) do not terminate with a poly(A) tail at the 3′ end, suggesting that H3.3 displacement might be central to carcinogenesis caused by polyadenylation of H 3.3 from active promoters, enhancers, and insulator regions.

Genetic ablation of Pim1 or pharmacologic inhibition with TP-3654 ameliorates myelofibrosis in murine models.
Avik Dutta,Dipmoy Nath,Yue Yang,Bao T. Le,Mohammad Ferdous-Ur Rahman,Patrick Faughnan,Zhenjia Wang,Matthew Stuver,Rongquan He,Wuwei Tan,Robert E. Hutchison,Jason M. Foulks,Steven L. Warner,Chongzhi Zang,Golam Mohi +14 more
TL;DR: In this paper, the authors showed that genetic ablation of Pim1 blocked the development of myelofibrosis induced by Jak2V617F and MPLW515L.

The ENCODE Imputation Challenge: a critical assessment of methods for cross-cell type imputation of epigenomic profiles
Jacob Schreiber,Carles Boix,Jin-Wook Lee,Hongyang Li,Yuanfang Guan,Jen-Chien Chang,Alex Hawkins-Hooker,Bernhard Schölkopf,Gabriele Schweikert,Mateo Rojas-Carulla,Arif Canakoglu,Francesco Guzzo,Luca Nanni,Marco Masseroli,Mark James Carman,Pietro Pinoli,Chenyang Hong,Kevin Y. Yip,Jeffrey P. Spence,Sanjit S. Batra,Jun S. Song,Shaun Mahony,Zheng Zhang,Wuwei Tan,Yang Shen,Yuanfei Sun,Minyi Shi,Jessika Adrian,Richard Sandstrom,Nina Farrell,Jessica Halow,Kristen Lee,Lixia Jiang,Xinqiong Yang,Charles B. Epstein,J. Seth Strattan,Michael Snyder,Manolis Kellis,William Noble,Anshul Kundaje +39 more
TL;DR: In this article , the authors comprehensively analyzed 23 methods from the ENCODE Imputation Challenge and found that imputation evaluations are challenging and confounded by distributional shifts from differences in data collection and processing over time, the amount of available data, and redundancy among performance measures.

TL;DR: By objective assessment of variant predictions, this work provides insights into current state-of-the-art algorithms and platforms for genome sequencing analysis for rare disease diagnosis and explore areas for future optimization.

Polyadenylation of Histone H3.1 mRNA Promotes Cell Transformation by Displacing H3.3 from Gene Regulatory Elements
Danqi Chen,Qiao Yi Chen,Zhenjia Wang,Yusha Zhu,Thomas Kluz,Wuwei Tan,Wuwei Tan,Jinquan Li,Jinquan Li,Feng Wu,Lei Fang,Lei Fang,Xiaoru Zhang,Rongquan He,Rongquan He,Steven S. Shen,Steven S. Shen,Hong Sun,Chongzhi Zang,Chunyuan Jin,Max Costa +20 more
TL;DR: Novel insights are provided into the importance of proper histone stoichiometry in maintaining genome integrity and the role of polyadenylation of canonical histone H3.3 at active promoters, enhancers, and insulator regions through its displacement are provided.
