Optimization of long-range PCR protocol to prepare filaggrin exon 3 libraries for PacBio long-read sequencing
Chiara Mareso,Elena Albion,William Cozza,Benedetta Tanzi,S. Cecchin,Paolo Gisondi,S Michelini,Francesco Bellinato,S Michelini,Silvia Michelini,Matteo Bertelli,Giuseppe Marceddu +11 more
TL;DR: In this paper , a 2-step long-range PCR protocol was used to generate 13-kb amplicons covering the whole filaggrin exon 3 sequence, which can be used for identification of LoF variants that are causative of the patients' disorders.
Towards a Long-Read Sequencing Approach for the Molecular Diagnosis of RPGRORF15 Genetic Variants
Gabriele Bonetti,William Cozza,A. Bernini,J. Kaftalli,Chiara Mareso,F. Cristofoli,Maria Chiara Medori,Leonardo Colombo,Salvatore Martella,G. Staurenghi,Anna P. Salvetti,Benedetto Falsini,Giorgio Placidi,Marcella Attanasio,Grazia Pertile,Mario Bengala,Francesca Bosello,Antonio Petracca,Fabiana D'Esposito,B. Toschi,Paolo Lanzetta,Federico Ricci,F. Viola,Giuseppe Marceddu,Matteo Bertelli +24 more
TL;DR: The results showed that NGS has a low coverage of the ORF15 region, while PacBio was able to sequence the region of interest and detect eight genetic variants, of which four are likely pathogenic, and provide a way to predict their pathogenicity.

Microbiome characterization of alpine water springs for human consumption reveals site- and usage-specific microbial signatures
Renato Pedron,Alfonso Esposito,William Cozza,Massimo Paolazzi,M. Cristofolini,Nicola Segata,Olivier Jousson +6 more
TL;DR: In this article , the authors used 16S rRNA high throughput sequencing to characterize the microbiome of 38 water springs in Trentino (Northern Italy) for two consecutive years in order to gain precious insights on the microbiome composition of these unexplored yet hardly exploited environments.
